Pedate pinellia Rhizome,the rhizome of Pinellia pedatisecta Schott which is classified into Areceae species,is a traditional Chinese medicine(TCM)that has long been used to cure thanatophidia bite,nameless swelling and toxicum,and cancer.In TCM books,it was recorded to possess efficacy in dispelling wind and relieving convulsion,drying dampness to eliminate phlegm,and eliminating stagnation.In TCM theory,expectorant play an important role in anti-aging.P.pedatisecta shows notable antiumor efficacy,and may anti-aging by its founction of expectorant.Up to the recent years,the relevant researches of P.pedatisecta are still not systematic and deep yet.In this study,PPE,a 95% ethyl alcohol extract obtained from the rhizomer of P.pedatisecta was evaluate by D-galactoseinduced subacute aging rats.Its activities of anti-aging and the possible mechanism were studied.Objective: By using D-galactose-induced subacute aging rats,Anti-aging activities of Ethanol extracts from Pinellia pedatisecta Schott were studied and the possible mechanism of the activities was discussed.Method: The aging rats were injected with D-gal.The Wistar rats in the treated group received different doses of PPE and rats in the control group were treated with 0.9% NaCl solution instead.The animals were weighted every four days.By using the Morris water maze test to detect the ability of learning and memory of the model rats.Relevant commercial kits were used to evaluate the activity of superoxide dismutase(SOD),the activity of telomerase(TE),Total antioxidantcat capacity(T-AOC),and the content of malonaldehyde(MDA)in brain and liver tissues.The expression of p53 and FOXO3 a in brain and liver were determined by immunohistochemical staining.Expression of the SIRT1,p53 and FOXO3 a gene in brain and liver tissues were measured by real-time RT-PCR.Western blot analysis was used to examin the expression of HuR,DBC1,p53 and FOXO3a protein.Results:(1)morphological analysis of all groups showed that the appetite,the coat color,behavior and mental condition of the treated groups and normal control were better than aging model.(2)Compared to the normal control,PPE had no significant effects on Body weight rate of the treated groups and aging model.(3)Morris water maze test data showed that:compared to the model group,the latency of escape and the total distance of exploration were significantly shortened.And the swimming speed was accelerated.(4)The activities of SOD and TAOC in the treated groups were significantly higher than those in normal controland model group(p<0.01).The contents of MDA in the brain and liver in treated groups were much less than that in the aging group.(5)PPE had an effect of increase theactivity of TE,and there was a significant difference compared to the aging model.(6)Compared to the aging model,the positive expression rate of p53 and FOXO3 a protein in normal control and treated groups were much higher,and the difference was significant(P<0.05).(7)The data showed that SIRT1,p53 and FOXO3 a mRNA expression were increased when compared to the aging group.(8)Western blot analysis showed that the expression of HuR,p53 and FOXO3 a protein in treated groups were higher than that in aging group,and the expression of DBC1 was lower.Conclusion: PPE could effectively attenuate the morphological damage in coat color,behavior and spirit induced by D-gal.In addition,it could significantly improve the ability of learning and memory of the model rats.Moreover,PPE could effectively enhance the the activity of SOD,T-AOC and TE,and decrease the MDA content in the treated groups.The date showed that PPE could Slow down the process of oxidative stress injury and has a great antioxidant activity.Its possible mechanism maybe concerned with the regulation expression of SIRT1,p53,FOXO3 a gene and HuR,DBC1,p53 and FOXO3 a protein.Overall,PPE is a potent natural anti-aging agent which can be developed as a new medicine and cosmetic for the treatment of age-related conditions.
