Objective:To investigate the effects and mechanism of anti-adhesion, anti-invasion and anti-metastasis of ginsenoside Rg3alone or combined with Oxaliplatin, Sorafenib on human hepatocellular carcinoma cell lines SMMC-7721in vitro,so as,to provide the theoretical foundation for the combination application of traditional medicine with chemotherapy and molecular targeted therapy to cure hepatocarcinoma.Method:To select human hepatocellular carcinoma cell lines SMMC-7721as the study object.We observe differences between drug groups and control group of human hepatocellular carcinoma cell lines SMMC-7721in vitro when they are affected by ginsenoside Rg3, Oxaliplatin,Sorafenib alone and by the combinative use of the drugs above by MTT method, cell adhesion experiment, cell migration experiment, cell invasive experiment and detecting the expression of apoptosis-associated gene protein of CD44V6,VEGF-C and MMP-9.Results:(1) MTT methods:Sorafenib (1.25μmol/L),ginsenoside Rg3(7.5μg/ml) and Oxaliplatin (0.25μg/ml) alone or by the combination after24hours can inhibit the proliferation of SMMC-7721cell lines,the apoptosis and proliferation rate is less than15%when we use each group of drugs above.(2)Cell adhesion experiment:The number of SMMC-7721cells adhesion to FN reduces to various degrees when the SMMC-7721cells are affected by each group of drugs above, this adhesion inhibition rate shows increasing trend depending on time when they are affected by ginsenoside Rg3(p<0.05); The adhesion inhibition rate in1hour shows no statistical significance (P>0.05). two-drug combination groups have better adhesion inhibition than single drug (Q>1.15);For2huors.the two-drug combination groups containing Sorafenib have better adhesion inhibition than Sorafenib single drug (P<0.05).but other two-drug combination groups do not show significant difference than single drug(P>0.05).The number of SMMC-7721cells adhesion to matrigel:the adhesion inhibition rate in1hour shows no statistical significance (p>0.05), two-drug combination groups have better adhesion inhibition than single drug (Q>1.15);For2huors.the two-drug combination groups containing Sorafenib have better adhesion inhibition than Sorafenib single drug (P<0.05).but other two-drug combination groups do not show significant difference than single drug (P>0.05);three-drug combination group has better adhesion inhibition than two-drug combination groups (P<0.05) when adhesion with matrigel.(3) Cell migration experiment:Sorafenib and Oxaliplatin single drug can reduce SMMC-7721cells in migration capacity. Two-drug combination groups are better than single drug (P<0.05). three-drug combination group is better than two-drug combination groups (P<0.05).(4) Cell invasive experiment:Sorafenib and Oxaliplatin single drug can reduce SMMC-7721cells in invasive capacity. Two-drug combination groups are better than single drug (P<0.05). Three-drug combination group is better than two-drug combination groups(P<0.05).(5) Detecting the expression of apoptosis-associated gene protein of CD44V6.VEGF-C and MMP-9:Ginsenoside Rg3can reduce the expression of CD44V6and MMP-9(P<0.05),however,can not affect the expression of VEGF-C. Oxaliplatin can reduce the expression of CD44V6^VEGF-C and MMP-9(P<0.05).Sorafenib can downregulate the VEGF-C and MMP-9expression of SMMC-7721cells (P<0.05),however,can not affect the expression of CD44V6. Two-drug combination groups are better than single drug (P<0.05). Three-drug combination group is better than two-drug combination groups (P<0.05).Conclusions:(1)Ginsenoside Rg3, sorafenib and Oxaliplatin single drug can reduce SMMC-7721cells in adhesion,migration and invasive capacity.In the adhesion aspect, Ginsenoside Rg3and Oxaliplatin can inhibit the adhesion capacity of SMMC-7721cells markedly. Two-drug combination groups have better adhesion inhibition than single drug. Three-drug combination group has better adhesion inhibition than two-drug combination groups. In the migration and invasive aspect, sorafenib and Oxaliplatin can inhibit the migration and invasive capacity of SMMC-7721cell markedly. Two-drug combination groups have better adhesion inhibition than single drug. Three-drug combination group has better migration and invasive inhibition than two-drug combination groups.(2)Ginsenoside Rg3can reduce the expression of CD44V6and MMP-9. Oxaliplatin can reduce the expression of CD44V6、VEGF-C and MMP-9.Sorafenib can downregulate the VEGF-C and MMP-9expression of SMMC-7721cell. Two-drug combination groups are better than single drug. Three-drug combination group is better than two-drug combination groups.
