AIMS:observe the effect of RNAi-mediated silencing of the hIAP-2gene on the proliferation and invasion of SMMC-7721cells.Methods:Three siRNAs toward hIAP-2gene were designed and transfected to HCC cell line SMMC-7721.Uuniversal scrambled negative control siRNA group (NC group). The gene expression of hIAP-2was determined using real-time quantitative PCR and western blotting. The cell viability was determined by MTT assay. The Annexin V-FITC/PI double-labeled flow cytometry was performed to measure the apoptosis at48h after transfection. Transwell migration assays were used to evaluate migration of transfected cells.Results:Real time PCR and western blotting test results showed that three hIAP-2-siRNAs could effectively inhibit hIAP-2mRNA and protein expression(P<0.05).Among them, hIAP-2-siRNAl silence has the most significant effect(P<0.05). MTT assay result displayed that the cell growth and proliferation were inhibited obviously for the knowdown group compared to control cells at24,48and72hours(p<0.05); Transwell test results showed that transfection hIAP-2-siRNA and untransfected negative control siRNA invasion irradiated group were significantly different(p<0.05);Conclusion:hIAP-2gene expression was efficiently silenced by hIAP-2siRNAs. Furthermore, knockdown of hIAP-2inhibits proliferation and invasion capability,and promotes apoptosis in HCC SMMC-7721cells.
