Background: Hyperuricemia is an important risk and predictive factor ofcardio-vascular and metabolic diseases, hyperuricemia will induced to endothelialdysfunction and many kinds of cardio-vascular diseases. Hyperuricemia is alsoassociated with the occur and development of many diseases such as insulin resistance、blood lipid disorder and renal disfunction. The conventional drugs in clinical, includingbenzbromarone, probenecid, allopurinol, etc.they have many side effects andunidentified therapeutic efficacy.In the past research,we had founded the micehyperuricemia model successfully by given a certain dose of Yeast+Ethambutol,andproved that the blood uric acid level of the mice hyperuricemia model could bedecreased sinificantlly by given vitamin B6、vitamin B12and folic acid,the protectiveeffect on blood vessel endothelium was also be observed.But the contrast about the uricacid decrease amplitude of mice hyperuricemia model by given a single vitamins B isnot in progress,it is also not very clear that which kind of vitamins B has the bettereffect on blood vessel endothelium protection.The Compound Danshen Dripping Pillhas a significant effect on many diseases including CHD、angina、hypertension、leftventricular hypertrophy、hyperlipemia and renal disfunction,but the concrete research isnot in progress about weather The Compound Danshen Dripping Pill has the effect ofdecreasing uric acid level or not,and its effect on blood vessel endothelium protection isalso not clear.Objective: The mice hyperuricemia model is founded by given Yeast+Etham-butol.The influence on uric acid level of the model is observed by administereddifferent kind of vitamins B and The Compound Danshen Dripping Pill,the effect ofeach group will be compared with others and the mechanism will be analysed,we hopethat the result could make a guidance on the clinical uric acid control treatment.At the same time, the effect on blood vessel endothelium protection by administered differentkind of vitamins B and The Compound Danshen Dripping Pill will also be explored andcompared,the mechanism about blood vessel endothelium disfunction which caused byuric acid will be discussed.Methods:90SPF-class male Kunming mice were randomly divided into7groupsaccording to body weight,12in control group(Yeast+Ethambutol+distilled water),13in allopurinol group(Yeast+Ethambutol+allopurinol),13in VB2group(Yeast+Ethambutol+VB2),13in VB6group(Yeast+Ethambutol+VB6),13in VB12group(Yeast+Ethambutol+VB12),13in folic acid group(Yeast+Ethambutol+folicacid),13in The Compound Danshen Dripping Pill group(Yeast+Ethambutol+TheCompound Danshen Dripping Pill). All the medications were administered for3weeks,then all mice were weighed,picked out eye balls to get blood,serum uric acid、serum NOand serum ET level of each group were measured.Separated mice thoracic and abdomi-nal aorta, sections for HE staining and detection of the expression of NF-κB andVCAM-1with immunohistochemical methods.Results:1、All the trial group(allopurinol、VB2、VB6、VB12、folic acid and TheCompound Danshen Dripping Pill) had a significant statistical disparity(P<0.01)compared with the control group on the serum uric acid level;There was no significantstatistical disparity(P>0.05)between allopurinol group and VB6、VB12、folic acidgroup;But there was a significant statistical disparity(P<0.01)between allopurinolgroup and VB2、The Compound Danshen Dripping Pill group;All the vitamins B grouphad a significant statistical disparity(P<0.01)compared with the Compound DanshenDripping Pill group.2、All the trial group had a significant statistical disparity(P<0.01)compared with the control group on the serum NO level; There was a significantstatistical disparity(P<0.01) between allopurinol group and VB2、 VB12、 TheCompound Danshen Dripping Pill group; But there was no significant statisticaldisparity(P>0.05)between allopurinol group and VB6、folic acid group; All thevitamins B group had a significant statistical disparity(P<0.01)compared with the Compound Danshen Dripping Pill group.3、All the trial group had no significantstatistical disparity(P>0.05)compared with the control group on the serum ET-1level;4、NF-κB of aorta endomembrane showed consecutive positive expression in thecontrol group,but NF-κB of aorta endomembrane in other groups were all attenuated invarying degrees.5、VCAM-1of aorta endomembrane showed consecutive positiveexpression in the control group, and VCAM-1of aorta endomembrane in other groupswere also attenuated in varying degrees.6、The morphous of aorta endomembrane andthe structure of vessel wall in each group were not changed during the trial period.Conclusion:1. The uric acid level of mice hyperuricemia model could be decreasedsignificantly by VB2、VB6、VB12、folic acid and The Compound Danshen Dripping Pill.Compared with allopurinol,VB6、VB12and folic acid almost had the same effect onuric acid level decrease, VB2maybe could not reach the effect of allopurinol, TheCompound Danshen Dripping Pill had a better effect than allopurinol and also betterthan other vitamins B.2. Hyperuricemia made lesion on blood vessel endothelium andthe serum NO level was decreased, VB2、VB6、VB12、folic acid and The CompoundDanshen Dripping Pill could increase serum NO level effectively and The CompoundDanshen Dripping Pill also had a better result.3.No statistical disparity were found onthe serum ET level variation which caused by vitamins B and The Compound DanshenDripping Pill,we thought it maybe associated with short trial period and(or)unsatisfactory medication dosage.4. Allopurinol、VB2、VB6、VB12、folic acid and TheCompound Danshen Dripping Pill could inhibit the expression of NF-κB and VCAM-1in mice aorta endomembrane in varying degrees.
