Objective: To investigate the expression of Bmi-1protein and p14ARF protein inhuman esophageal squamous cell carcinoma and normal esophageal mucosa tissue aswell as their correlations with the clinicopathologic factors of patients with esophagealsquamous cell carcinoma.Methods:1. the expression of Bmi-1protein and p14ARF protein in49cases ofesophageal squamous cell carcinoma (ESCC) and27cases of normal esophagealmucosa tissue was detected by Elivision method of immunohistochemical staining andits relationship to clinicopathological factors was analyzed. Then to analyse thecorrelations of Bmi-1protein and p14ARF protein in human esophageal squamouscell carcinoma.2. We make the cases of Bmi-1protein expresson is negative and p14ARF proteinexpresson is positive as favourable expression group, the cases of Bmi-1proteinexpresson is positive and p14ARF protein expresson is negative as unfavourableexpression group.So37ESCC cases were divided into favourable expression group andunfavourable expression group,and then to analyse the relationships of they withclinicopathological factors of ESCC.3. Statistical analysis was performed by spss13.0for windows. All tests was conductedat a significance level ofα=0.05.Results:1. The positive expression rate of Bmi-1protein in ESCC and normalesophageal mucosa tissue was79.59%(39/49) and11.11%(3/27) respectively (P<0.01).The positive expression rate of p14ARF protein in ESCC and normal esophagealmucosa tissue was28.57%(14/49) and74.07%(20/27) respectively (P<0.01).2. The positive expression of Bmi-1protein was not related to age and gender ofpatients, histological differentiation,length of tumor,location, depth of invasion, lymphnode metastasis and clinical stage (P>0.05)in49cases of ESCC.3. The positive expression of p14ARF protein was not related to age and gender ofpatients, histological differentiation,length of tumor,location,depth of invasion, lymphnode metastasis and clinical stage (P>0.05)in49cases of ESCC.4. There was a negative correlation between the positive expression of Bmi-1protein and p14ARF protein in ESCC (rs=-0.352, P=0.013).5. There is no significant differrence between the favourable expression group andunfavourable expression group in gender and age of patients,histological differentiation,length of tumor,location, depth of invasion, lymph node metastasis and clinical stage (P>0.05)in37cases of ESCC.Conlusion:1. The expression of Bmi-1protein and p14ARF protein in ESCC isabnormal.Bmi-1and p14ARF may play an important role in the carcinogenesis ofESCC.2. The expression level of Bmi-1protein and p14ARF protein did not correlate with anyclinicopathologic factors.3. There was a negative correlation between the abnormal expression rate of Bmi-1protein and p14ARF protein in ESCC,this suggests that Bmi-1and p14ARF make asynergistic action in the carcinogenesis of ESCC,Bmi-1—p14ARF pathway play animportant role in the carcinogenesis of ESCC.4. The combined detection of Bmi-1protein and p14ARF protein are not a valuablemarker of ESCC progression,Bmi-1and p14ARF can be a candidate biomarker for thedevelopment of diagnostic.
