Objective: Type2diabetes is caused by a variety of causes, metabolicdisorders characterized by chronic hyperglycemia associated withinsufficient insulin secretion and/or insulin resistance caused by thesugar, fat and protein metabolism abnormalities. In order to improve thequality of diabetic patients’ life, many complications of diabetes, diabeticulcers, gangrene, local infection, trauma refractory become the focus oftoday’s medical research. At present, medical biological model of woundhealing for studies are mainly rodents. Study found that rodents traumahealing process mainly through the wound contraction, and humanwound healing by granulation tissue formation and regeneration ofepidermal cells to complete, therefore, both the healing process of somedifferences, thus not the true reaction of human wound healing process.Therefore, in order to avoid the trauma of rodents wound contractionafter the impact to the experimental, the experimental application ofsilicone resin fixed wound around the skin, minimizes wound contraction, so this process to better reflect human wound healing process. Modelbasis for the study of diabetes difficult wounds refractory.Methods: Type2diabetes was induced by combination of high-fatdiet-fed and low-dose streptozotocin (STZ,45mg kg-1,ip) injection in SDrats, make a circular wound with the punch on the dorsum of the SDmice, The silicone splint was used to fix the bound of wound followed byinterrupted6–0nylon sutures to ensure position, Minimizes woundcontraction, the simulation of human wound healing process.In differentsensible point to take the wound granulation tissue around the biopsywound margin of granulation tissue in skin thickness, microvesseldensity, collagen fiber and collagen content changes.Results: Compared with normal group, after72h STZ injection, ratsfed with high fat whose fasting blood glucose(22.71±7.26;P<0.01),triglycerides(4.54±0.96;P<0.01)and cholestero(l1.91±0.08;P<0.05)significantly increased, the differences were statistically significant;Compared with diabetes with silicon ring not fixed group, after a traumaof1,3,5,7,14days,the skin thickness, microvascular density,collagenfibers and collagen content of diabetes group were significantly changed.Conclusions:(1)High-fat diet and STZ induced type2diabetes issimple and effective.(2)Use silicon ring fixed and the effect flange toslow down the wound shrinkage speed.(3)Silicon ring fixed wound can effectively increase the granulation tissue forming and epidermal cellsregeneration.
