Objective:To observe the influence of Cromoglycate disodium on the expressionof chymase、IL-6、bFGF and the formation of neovascularization in the Wistar rat’satherosclerotic plaque, and research the role of mast cell in the destabilition ofatherosclerotic plaque in the wistar rat.Method:The36male Wistar rats (7-8weeks old) were randomly divided into3groups and12rats in each group. Group A were treated with high-fat (+PTU) feedingand VitD3+OVA injection;Group B were treated with injection Disodium Cromoglycateon the basis of group A. Group C were treated with high-fat (+PTU) feeding and VitD3injection. All wistar rats were fed a high-cholesterol diet for8weeks. At the beging ofweek9,the rat of group A were challenged intravenously by injection of albumin(VOA)toinduce mast cell activation for3weeks before euthanasia, while the rat of group Breceived an intraperitoneal injection of the mast cell stabilizer cromolyn before and afterOVA challenge. After12weeks, extracting the aortic, HE staining was used to observedplaque pathology morphology. Immunohistochemistry was used to measure theexpression of CD31and RT-PCR was used to measure Chymase, bFGF mRNAexpression in the rats’ atherosclerotic plaques.Results:1. A, B, C three groups formed arterial atherosclerotic,in which GroupA,atherosclerosis pathological changes were significantly heavier than the Group B andC, a large number of foam cell formation in plaque,at the bottom of plaque the foam celldegeneration and necrosis,atherosclerotic plaque broke into the lumen obviously; Plaque area of group A was significantly increased compared witht the groups B, C (group A73000±9000μm~2vs group B43000±7000μm~2P<0.05group A73000±9000μm~2vsgroup C37000±9000μm~2P<0.05);2.Immunohistochemistry showed there werepositive CD3l expressions in the plaques.The protein optical density values (IOD) ofCD3l in group A are significantly higher than group B and C;3.According to RT-PCRanalysis, the relatively gray-scale value of Chymase、IL-6、bFGF mRNA in group A aresignificantly higher than group B and C.Conclusion:1. Cromoglycate disodium make the expression of chymase, IL-6bFGF and the formation of neovascularization in the Wistar rat’s atherosclerotic plaquesignificantly reduced2. Mast cells membrane stabilizer may inhibit the AS developmentand reduce plaque instability3.Mast cells play a crucial role in atherosclerotic plaqueprogression and destabilization in the Wistar Rat.
