Objective:To evaluate the efficacy of the transplantation of BMSCs combinated withVEGF with the transplantation of BMSCs or VEGF in the treatment of middlecerebral artery ischemia (MCAO) models.We investigated the effect of BMSCscombinated with VEGF on promoting functional improvement for treatment of rat’scerebral ischemia injury and its possible mechanism on the infarction volume, thescores of neurologic function and the expression of MAP2,.Methods:We gain BMSCs by all cellulae medullares and adherent method. Rat middlecerebral artery occlusion (MCAO) model was established by improved method.TheSD rats that have arrived at the scores(n=48) were randomly divided into PBSgroup(B), BMSCs group(C),VEGF group(D) and BMSCs with VEGF group(E) atrandom. The sham group A(n=12), only isolated to the carotid artery and sutured theskin without any treated. After24hours the MCAO models were establishedsuccessfully, Group C and E were transplanted intravenously with2×106/L BMSCssuspensions by the caudal vein respectively.Group B received PBS. And after2daystransient MCAO, group D and E were given the VEGF intranasally. The neurologicalscore was estimated after24hours and on day14. Rats were euthanized on day14,thebrain sections were observed by TTC staining and an image analysis system was usedto caiculate the infact volume. HE staining was used to observe the morphology.Theimmunoreactivity of MAP2was determined by using immunohistochemical stainingand computer image analysis system. Results:1. The scores of neurologic function: The sham operated group was the same asnormal SD rats. Neurological function score show that the groupPBS, BMSCs, VEGFand BMSCs with VEGF were3.41±0.51,2.58±0.51,2.67±0.47,1.91±0.46,respectively. Group PBS of the scores of neurologic function was marklydifferent to the other groups(P<0.05). The score of the group BMSCs with VEGF wassignificantly fewer than that in the other groups(P<0.05).2. HE stain: With low power lens, in the PBS group, the brain tissue wassignificantly edematous, vacuolar degeneration and the space of cells became larger,the volume of nerve cells was increased. The BMSCs with VEGF group had morenerve cells and abundant cytoplasm. The cell damage was the lowest in all groups.Some small softening focus was scattered, the other structure was better.3. Infarction volume: The infarction volume show that the group sham oprated,PBS, BMSCs, VEGF and BMSCs with VEGF were0mm3,239.58±6.89mm3,188.83±10.231mm3,186.58±8.45mm3,126.92±12mm3. The average infarct volumeswere significantly reduced in BMSCs and VEGFgroups compared with those of thePBS group(P<0.05). The infarct volume was significantly decreased in the BMSCswith VEGF group as compared with the other groups(P<0.05).There was nosignificant difference found between the infact volumes of the BMSCs and the VEGFgroup (P>0.05).4. Immuohistochmistry:The expression level of MAP-2of group A,B,C,D,Ewere:0.701±0.014,0.413±0.038,0.419±0.019,0.614±0.035.Group B was the lowest,compared with the other groups(P<0.05).Group E showed a statistically significantdifference compared with group C and group D(P<0.05).Conclusion::1. Bone marrow stromal cells(BMSCs) transplantation combined with vascularendothelial growth factor (VEGF) group,s therapeutic efficacy is more obvious thanthat’s of BMSCs and VEGF group.2. Increasing the expression of MAP2induced by cerebral ischemia may be oneof the mechanisms that the BMSCs transplantation combined with VEGF can improve neurologic function.
