Relationship Between The Levels Of LCAT And Cetp And The Distribution Of Plasma Hdl Subclasses In Patients With Coronary Heart Disease

Objective: To study the relationship between the levels of lecithin cholesterolacyltransferase (LCAT) and cholesteryl ester transfer protein (CETP) and the contentsof plasma high-density lipoprotein (HDL) subclasses in patients with coronary heartdisease (CHD).Methods: The contents of plasma HDL subclasses in healthy controls (n=89) andthe CHD subjects (n=89) were determined by two-dimensional gel electrophoresisassociated with immunodetection method. The contents of total cholesterol (TC),triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-densitylipoprotein cholesterol (LDL-C), apolipoprotein A1(apoA1) and apolipoprotein B100(apoB100) were detected by Automatic biochemical analysis instrument. The levelsof LCAT and CETP were determined by enzyme-linked immunosorbent assay(ELISA) kits.Results: Compared with control group, the contents of TC, TG, LDL-C, apoB100and CETP in CHD groups were increase significantly (P<0.05or P<0.001), while thelevels of apoA1, HDL-C, LCAT and the ratio of apoA1/B100were decreasesignificantly (P<0.001). The contents of preβ1-HDL, HDL3cand HDL3bin CHDsubjects were increase significantly (P<0.01or P<0.001), while that of HDL2a,HDL2bwere decrease significantly (P<0.001). The CHD subjects were stratifiedaccording to quartiles of the plasma LCAT levels (groupⅠ: LCAT <18.60μg/ml,group Ⅱ:18.60≤LCAT <33.00μg/ml, group Ⅲ:33.00≤LCAT <49.00μg/ml,group IV: LCAT≥49.00μg/ml). With the decrease of LCAT concentration, thecontents of HDL2bwere decrease significantly. Compared with group IV, the contentsof HDL2bin groupⅠand group II were significantly decrease (P<0.01). The CHDsubjects were stratified according to quartiles of the plasma CETP levels (groupⅠ: CETP <0.63μg/ml, groupⅡ:0.63≤CETP <0.84μg/ml, group Ⅲ:0.84≤CETP <1.13μg/ml, group IV: CETP≥1.13μg/ml). With the increase of CETP concentration,the contents of preβ1-HDL were increase significantly, while that of HDL2bweredecrease significantly. Compared with groupⅠ, the contents of HDL3cwere increasesignificantly in group Ⅲ (P<0.05). The contents of HDL2bin group Ⅳ weresignificantly higher than groupⅠ (P<0.05). The plasma CETP was positivelycorrelated with the contents of preβ1-HDL (r=0.228, P<0.01) and negativelycorrelated with the contents of HDL2b(r=-0.200, P<0.01). LCAT was positivelycorrelated with the contents of HDL2b(r=0.204, P<0.05).Conclusions: The particle size of HDL in CHD subjects showed a shift towardsmaller size in CHD subjects, and the tendency was related with the decrease ofLCAT levels and the increase of CETP levels.
