Objective: The tissue type, differentiation degree and Metastasis of gastriccarcinoma were closely related with the expression of different its genes, which it wasimportant for evaluating and predicting the prognosis of gastric carcinoma patients.This study aimed to analyze the expression of CK-8and P16proteins, screen therelative protein of gastric carcinoma, and provide valuable for clinical early detectionand evaluation clinical result of gastric carcinoma patients.Methods: The normal gastric mucosa, paracarcinoma, atypical hyperplasia, gastriccarcinoma and lymphonode metastatic carcinoma were respectively collected.Meanwhile, the total RNA of different tissues was extracted and the expression ofCK-8and P16genes was detected by RT-PCR. At the same time, the total protein ofdifferent tissues was extracted, and then the expression of CK-8and P16proteins wasexamined with western blot. The expression of CK-8and P16proteins was analyzedby immunohistochemistry with tissue array that included of normal gastric mucosa,paracarcinomas, atypical hyperplasia, gastric carcinoma and lymphonode metastaticcarcinoma tissue. The expression of two proteins and relationship were evaluated withclinical pathology in gastric carcinoma.Results:1. The result of RT-PCR showed that the expression of CK-8gene wasup-regulation in gastric carcinoma and lymphonode metastatic carcinoma, comparedwith normal gastric mucosa, carcinoma side tissue and atypical hyperplasia tissue(P<0.01). However, P16gene was down-regulation (P<0.01). Moreover, theexpression of CK-8gene in atypical hyperplasia tissue was higher than that in normalgastric mucosa, paracarcinoma (P<0.01), but P16gene was lower (P<0.01).2. The result of western blot displayed that the expression of CK-8protein was up-regulationin gastric carcinoma and lymphonode metastatic carcinoma, compared with normalgastric mucosa, paracarcinoma and atypical hyperplasia(P<0.01). However, P16protein was down-regulation (P<0.01). Moreover, the expression of CK-8protein inatypical hyperplasia was higher, but P16protein was lower than that in normal gastricmucosa, paracarcinoma (P<0.01).3. The consequence of tissue array stained byimmunohistochemistry exhibited that the expression rate of CK-8protein was11.76%(4/34),14.29%(7/49),43.31%(11/26),82.72%(67/81) and89.09%(49/55)respectively in normal gastric mucosa, paracarcinoma, atypical hyperplasia tissue,gastric carcinoma and lymphonode metastatic carcinoma. The positive rate of CK-8protein in gastric carcinoma and lymphonode metastatic carcinoma was higher thanthat in normal gastric mucosa, paracarcinoma and atypical hyperplasia tissue and thisdifference was statistical significance (P<0.01). The positive rate of CK-8protein inatypical hyperplasia tissue was stronger than that in normal gastric mucosa, and thevariance was statistical significance (P<0.01). Moreover, the expression rate of CK-8protein was60%(3/5),76.47%(13/17) and84.75%(51/59) respectively in gastricwell differentiated adenocarcinoma, moderately differentiated adenocarcinoma andpoorly differentiated adenocarcinoma. The expression of CK-8protein was graduallyadvanced with differentiation degree decreased, and the difference of groups wasstatistical significance (P<0.01). The expression rate of P16protein was76.47%(26/34),79.59%(39/49),34.62%(9/26),8.64%(7/81) and1.69%(1/55) respectivelyin normal gastric mucosa, paracarcinoma, atypical hyperplasia tissue, gastriccarcinoma and lymphonode metastatic carcinoma. The positive rate of P16protein ingastric carcinoma and lymphonode metastatic carcinoma was higher than that innormal gastric mucosa, paracarcinoma and atypical hyperplasia and this differencewas statistical significance (P<0.01). The positive rate of P16protein in atypicalhyperplasia tissue was stronger than that in normal gastric mucosa, and the variancewas statistical significance (P<0.01). Moreover, the expression rate of P16proteinwas20%(1/5),11.76%(2/17) and6.78%(4/59) respectively in gastric welldifferentiated adenocarcinoma, moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma. The expression of P16protein was gradually advancedwith differentiation degree decreased, and the difference of groups was statisticalsignificance (P<0.01).Conclusion:1. The expression of CK-8protein was up-regualtion in gastric cancer andlymphonode metastatic carcinoma compared with in normal gastric mucosa,paracarcinoma and atypical hyperplasia tissue, but the expression of P16proteinwas down-regualtion. The expression of CK-8protein in atypical hyperplasiatissue was stronger than that in normal gastric mucosa. However, P16protein waslower.2. The CK-8and P16proteins were related with occurrence, development, anddifferentaiation degree and lymphaden metastasis of gastric carcinoma.
