Objective Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common monogenetic inherited renal disease, estimated to account for about5-10%of end stage renal disease patients The aim of the study is to determine the expression of connective tissue growth factor (CTGF) and transforming growth factor-β1(TGF-β1) in ADPKD renal tissues, and to interpret the effect of CTGF and TGF-β1on the pathological mechanism and progression of ADPKD.Methods67ADPKD patients were chosen and divided into different groups. According to the glomerular filtration rate(GFR) of operative side kidney,67patients were divided into group A (50ml/min>GFR≥25ml/min) and B (25ml/min> GFR>10ml/min). According to the kidney volume of operative side kidney, they were divided into group1(kidney volume≤750cm3) and2(kidney volume>750cm3). According to the blood pressure of ADPKD patients, they were divided into group a (systolic pressure<160mmHg and diastolic pressure<100mmHg) and b (systolic pressure≥160mmHg or diastolic pressure≥100mmHg).24cases of normal renal tissue were chosen as control group. The expression of CTGF and TGF-β1were assayed by means of immunohistochemical technique with SABC method.Results (1) The positive expression rate of CTGF and TGF-(31in ADPKD was86.6%and83.6%respectively, which was significantly higher than that of the controls (P<0.05). Expression of CTGF was closely related to that of TGF-β1(P<0.05). The positive expression of CTGF in group A with lower GFR was higher than group B. The positive expression of CTGF in group1with higher kidney volume was higher than group2. The positive expression of CTGF in group b with higher blood pressure was higher than group a.Conclusion CTGF and TGF-β1simultaneously promote the progression of ADPKD. The level of CTGF significantly increased with the decrease of GFR and the increase of kidney volume and the increase of ADPKD patients’BP, which suggests CTGF might play an important role in the pathogenesis of cyst formation and interstitial fibrosis in ADPKD.
