Objective:To observe the renal interstitial fibrosis factor expression influence of ChongCaoYiShen Granules on rat model which is renal interstitial fibrosis with, and explore its mechanism of delay the progress of chronic renal failure and anti-renal interstitial fibrosis.Method:Male180SpragueDawley rats were randomly divided into four group: sham operation group, model group, fosinopril group, ChongCaoYiShen group. All rats except those from sham operation group received5/6nephrectomy to establish RIF model. Administration:intragastric administration were performed after model-establishing, and continued until12weeks. Sham operation group and model group received distilled water of the same amont, ChongCaoYiShen group received water decoction of ChongCaoYiShen Granules, and fosinopril group fosinopril sodium chloride solution once a day.24-hour urine protein quantity, BuN, Scr, Ang II were examined0,4,8,12weeks after model-establishing.10rats in each group were sacrificed at one time to observe histomorphological changes of kidney and expressions of TGF-β1, PDGF-BB, HGF and BMP-7.Results:(1)ChongCaoYiShen Granules may significantly reduce expression of BUN, Scr, Ang II in serum of rat, and lower urine protein comparing to sham operation group(P<0.01, P<0.05);(2)Using ChongCaoYiShen Granules and fosinopril to treat, the pathological changes of tubulointerstitial inflammatory cells infiltration, renal tubular atrophy and renal interstitial fibrosis to a lesser degree, ChongCaoYiShen group superior to fosinopril group;(3)ChongCaoYiShen Granules may significantly inhibit expression of TGF-β1、PDGF-BB comparing to sham operation group(P<0.01), and superior to fosinopril group(P<0.01,P<0.05);(4)ChongCaoYiShen Granules may significantly increase expression of HGF,BMP-7comparing’to sham operation group(P<0.01), and superior to fosinopril group(P<0.01,P<0.05).Conclusion:(1)ChongCaoYiShen Granules may regulate RIF rat whole body life quality, and delay disease development.(2)ChongCaoYiShen Granules may reduce content of BUN, Scr, Ang Ⅱ in serum of rat with RIF, lower urinary protein, therefore relieve RIF;(3) ChongCaoYiShen Granules may reduce renal interstitial fibrosis of5/6nephrectomy rat(4)ChongCaoYiShen Granules may inhibit the expression of TGF-β1,PDGF, increase the expression of BMP-7,HGF, relieve renal interstitial fibrosis progress effectively.
