Objective: To investigate whether Shengjingsan recipe has a therapeutic effect on ratmodel of asthenozoospermia by upregulation of CatSper channel in the sperm.Methods:(1)A rat model of asthenozoospermia was induced by intraperitonealinjection of cyclophosphamide (CTX) everyday lasted for14days. The sperm viability andthe sperm vitality, sperm concentration as well as the sperm mobility were examined usingcomputer assisted sperm analysis (CASA).(2) Expressions of CatSper1, CatSper2,CatSper3protein in the rat sperm of each group were measured using Western blotanalysis.The main results:(1) The parameters of the sperm viability, the sperm vitality andthe sperm concentration decreased significantly in CTX-treated rats as compared withna ve rats or normal saline-treated rats, indicating that CTX could induce typicalasthenozoospermia in rats.(2) The above decreased parameters of the sperm viability andthe sperm vitality, sperm concentration as well as the sperm mobility, induced by CTXtreatment, could be reversed effectively after Shengjingsan recipe treatment.(3) CTXsignificantly decreased the expression of CatSper in the rat sperm, whereas Shengjingsanrecipe could inhibit the CTX-induced decrease in the CatSper expression.Conclusions: Shengjingsan recipe has a therapeutic effect on asthenozoospermia byupregulation of CatSper expression in rat the sperm. |
