Objective:To investigate the effects of Chaihu Shugan San (CHSGS) on behavior and the expressing of BDNF and its receptor TrkB in the hippocampus, amygdala and frontal lobe of chronic mild unpredicted stress depression rats.Methods:Sixty adult Sprague-Dawley rats were randomly divided into6groups:normal control group (NC), model control group (MC), Chai-hu Shu-gan San group (CHSGS), disassembly1group (C Ⅰ), disassembly2group (CⅡ) and Fluoxetine control group (FC). All rats except the normal control group were singly housed and exposed on an unpredicted sequence of mild stressor for continuous4weeks. Since the fifteenth day, all the groups were intragastric administration, and respectively for equal volume normal sodium (NC and MC group), CHSGS, CⅠ, CⅡ and fluoxetine for2weeks. The behavior of rats was detected by open-field test and sucrose preference test on the0th,7th,14th,21th,28th day. The expression of BDNF and TrkB in hippocampus, amygdala and frontal lobe in different groups were detected by immunohistochemistry technique and Real-time fluorescent quantitation PCR.Results:Compared with NC group, the rats in model group showed significantly decreased body weight. Scores of crossing, rears and grooming in open-field test reduced significantly. Pure water consumption in sucrose preference test increased significantly. The expression of BDNF and TrkB in hippocampus, amygdala and frontal lobe decreased significantly. The depressive behavior of the rats were improved significantly and the expression of BDNF and TrkB in hippocampus, amygdala and frontal lobe were obviously increased in CHSGS and FC group.Conclusion:Chronic mild unpredicted mild stress can affect rats behavior, cause depression. Chai-hu Shu-gan San can obviously improve depressive state of the rats, its mechanism maybe was by increasing the expression of BDNF and TrkB in hippocampus, amygdala and frontal lobe.
