Backgroud and Objective:Alzheimer disease(AD) is a kind of cerebral degenerative disease menifestied by progressive amnesia and demensia which bring about incubus to society and family. Therefore medicine figure proceed deepgoing research for this. For the past fe w years chinese herbal drugs had acquired better therapeutic effect in treating AD. Compound Recipe Radix Polygoni Multiflori Extract (CRPME) was made up with formula on predecessor investigative basis, with invigorating the kidney activating blood circulation and espelling phlegm as its Therapeutic Principle, in this study we observe its clinic curative effect in120AD patients, the results shows that the total cerebral effective power is93.33%in CRPME, there was significant difference (P<0.05) compared with western medicine and single Chinese herbal drug Radix Polygoni Multiflori. CRPME group can raise MMSE score and decrease ADL score, which illustrating that it can all-rightly ameliorate AD patients’memory and conignitive ability, improving daily life and social activities ability, it is thus clear that CRPME group has already acquire better effect in clinical trials。With regard to the possible mechanism of action of CRPME treating AD has no empirical study yet, we investigate the action of CRPME on AD rats’cognitive and memory ability and the expression of (brain derived neurotrophic factor, BDNF) and (Tyrosine kinase receptor,TrkB)in the hippocampus of AD rats.The study discusses partially the mechanism of traditional Chinese medicine CRPME with respect to its effectiveness to treat Alzheimer’s disease And provide clinic evidence for treating AD.Methods:40male AD rats aged6-months were randomly divided into4groups, the normal group(group A),sham-operation group (group B),model group(group C), and the CRPME group (group D). Then the last two groups were injected by Aβ25-35in both sides of hippocampus to make AD models. Rats of group B were injected by the same volume of NS in the same way. NO treatments were done to group A. then test weather the AD model is successful or not with Morris water maze test (MWM). After that Rats of group D were respectively given CRPME,while the rest groups were given double-distilled water for8weeks. All the groups were tested with Morris water maze test (MWM) after medical treatment to test the change of AD patients’memory and leaning ability. Finally, the hippocampus were taken out to examine the expression of neurotrophic factor, BDNF) and (Tyrosine kinase receptor,TrkB) by semi-quantitative RT-PCR.Results:1.Behavioral results:In hidden platform experiment and exploratory experiment, compared with group A, the group C and D rats’ cognitive and memory ability delayed significantly (the escaped and latent period is extended, the average pass through times is decreased)After Chinese medicine treatment, Compared to group C, cognitive and memory ability of group D rats were strengthened significantly (the escaped and latent period is shortened, the average pass through times is increased)(P<0.05).2.RT-PCR results:Compared to group A, the expression of BDNF and TrkB in group C decreased (P<0.05), that in group B had no statistically significance.Compared to group C, the expression of BDNF and TrkB in group D increased (P<0.05).Conclusion: 1. By injecting condensed Aβ25-35to both sides of rats’ hippocampus can make representative AD model rats.2. Invigorating the kidney activating blood circulation and prescription CRPME can remarkably improve AD rats’cognition and memory ability.3. The decreased level of BDNF and TrkB in the AD rats’ hippocampus is probably part of pathology of Alzheimer’s disease.4. Invigorating the kidney activating blood circulation prescription CRPME can increase the expression of BDNF and TrkB in AD rats’ hippocampus, which probably accomplish its curative effects through ameliorating cholinergic nerve system and protecting neurons...
