Objective:This study is aimed to research the protective effect of Erigeron breviscapus against hepatic ischemia reperfusion injury and investigate it’s possible mechanism in rats.Methods:72adult male rat were randomly divided into the following4groups:group A, sham-operation group; Group B, saline control group; Group C, low-dose (5mg/kg) Erigeron breviscapus treatment group; Group D, high-dose (10mg/kg) Erigeron breviscapus treatment group.18rats in each group, the left-median liver lobe was blocked by a clamp for45minutes, the Erigeron breviscapus was injected though dorsal vein at1hour before operation and immediately postoperation,6rats in each groups were executed at2h,6h and24h postoperation. Blood samples were obtained to measure the levels of ALT and AST. Liver tissues were collected at the same time to measure the contents of protein MDA, SOD and to observe pathologic histology, the apoptosis of hepatocytes were detected by TUNEL, the levels of Bcl-2and Bax was measured by immunohistochemistry.Results:1. Compared with group B, group C and group D:The blood serum levels of ALT, AST were significantly decreased(P<0.05); The level of SOD in liver was significantly increased(P<0.05) and the level of MDA was significantly decreased(P<0.05); The cell apoptosis index were significantly decreased(P<0.05); The expressions of Bcl-2in liver were significantly increased, in addition, the expression of Bax were significantly decreased(P<0.05).2. Compared with group C, group D:The blood serum levels of ALT, AST were significantly decreased(P<0.05); The level of SOD in liver was significantly increased(P<0.05) and the level of MDA was significantly decreased(P<0.05); The cell apoptosis index were significantly decreased(P<0.05); The expressions of Bcl-2in liver were significantly increased, in addition, the expression of Bax were significantly decreased(P<0.05).Conclusion:1. Erigeron breviscapus can prevent liver against ischemia reperfusion injury which may associate with the reducing of MDA and the improvment of SOD.2. The inhibitation of hepatocytes apoptosis possibility related to the decrease of Bax and the increase of Bcl-2in liver tissue, which protect liver agaist ischemia reperfusion injury.
