Objective: We aim to investigate erigeron breviscapus and its active component of anti-tumor effect and their molecular mechanisms.Methods: Cell culture in vitro and capillary-like networks structure experiments were used to observe a variety of Chinese herbal erigeron breviscapus extracts of anti-tumor effect. Alamar Blue assay was used to determine the active component of erigeron breviscapus - scutellarin on 38 kinds of tumor cell lines proliferation. Thus we found this active component scutellarin had a strong inhibition on human Burkitt's lymphoma cell line Namalwa proliferation. The change of cell cycle status in Namalwa cells treated with different concentration scutellarin was performed by the flow cytometry. Annexin V-FITC and PI staining was used to detect the effect of scutellarin on Namalwa cells apoptosis. The mRNA expression of genes related to cell cycle regulation was detected by RT-PCR. The altered expression level of several apoptosis-associated proteins induced by scutellarin was detected by western blot.Results: Chinese herbal erigeron breviscapus extracts can effectively inhibit capillary-like networks structure. The active component of erigeron breviscapus- scutellarin has good anti-tumor effect. Scutellarin exhibited potent inhibition activity against the growth of 38 kinds of tumor cell lines especially it could strongly inhibit Namalwa cells proliferation. The inhibition ratio increased with scutellarin concentration increased. The distribution ratios of cells were significantly different compared with those of the controls (P<0.05). Flow cytometry analysis showed that the cell cycle phage distribution changed. With scutellarin concentration increased, accumulation of the cells in the G0/G1 phase arrest, G2/M phase of the cell cycles decline after Namalwa cells treated with scutellarin. It was significantly different compared with those of the controls(P<0.05). Annexin V-FITC and PI staining confirmed that scutellarin could induce Namalwa cells apoptosis. RT-PCR showed the correspondent expression of cyclinD1 and CDK4 mRNA was significantly decreased. Western blots showed when Namalwa cells treated with scutellarin a significant decrease in caspase3 and cytochrome c, and a significant increase in cytochrome c polymers levels.Conclusion: Chinese herbal erigeron breviscapus can inhibit the growth of tumor cell lines. While its active component -scutellarin has good anti-tumor effect especially on lymphoma cell line Namalwa. Scutellarin can inhibit Namalwa cells proliferation via regulation of cyclinD1 and CDK4 expression in low dose, and in high concentration it can induce Namalwa cells apoptosis altered proteins expression level of caspase3 and cytochrome c.
