Objective:This research focus on the express of Hepatocyte growth factor and its receptor c-Met in primary hepatocellular carcinoma.Methods:31patients of primary hepatocellular carcinoma were choosed to be experiment group,and there were31patients of every group with Decompensated liver cirrhosis,Mild chronic hepatitis B patients and normal liver function in patients with gallbladder polyps were choosed to be control groups.Using enzyme immunoassay (ELISA) quantitative method to determination the experimental group that one day before operation, and after the fourth.the ninth.the eleventh days of serum HGF concentration.Using immunohistochemical analysis and Reverse transcriptase polymerase chain reaction(RT-PCR) to detection of carcinoma, paracarcinoma and normal liver tissue in c-met protein expression.The same method the concentrations of serum HGF detection control group.At the same time analysis of the relationship between the preoperative HGF level in serum and cancer tissue in the expression of c-met degree with different clinicopathological factors and influence the factor that the changes of the serum level of HGF after operation.Result:PLC patients with decompensated liver cirrhosis patients with serum HGF concentration is higher than the normal control group and the group of chronic hepatitis B,the difference has statistically significant;PLC patients with decompensated liver cirrhosis patients between serum HGF concentrations of no significant differences;Postoperative HGF concentration increase peak appeared on the fourth day,After ninth, eleventh days decreased gradually.From the preoperative to postoperative day fourth,Resection of large section increased more obviously than segment resection in serum HGF concentrations(P=0.021); Expression of c-met protein in carcinoma tissue intensity was significantly higher than that of adjacent tissues and normal tissues,But in adjacent carcinoma tissue,The expression of c-met protein in liver cirrhosis group higher than in those without liver cirrhosis group(P=0.026); C-met protein expression in cancer tissue and serum HGF concentration was positively correlated with the intensity of (R=0.612); PLC patients preoperative serum HGF levels was correlated to tumor size and There is no complicated with liver cirrhosis and There are no relevant to portal vein tumor thrombi; Compared with the preoperative liver function, serum AFP levels without correlation.Cancer of c-met protein expression in strength with and without combined with portal vein tumor thrombus associated,And there is no correlation with the size of the tumor, the serum AFP level, and there are no cirrhosis of the liver; The case of Postoperative tumor recurrence or metastasis,preoperative serum HGF level and c-Met protein expression in tumor tissues was significantly higher than that of no recurrence or metastasis.Conclusion:The presence of serum HGF in patients with PLC and the expression of c-met protein in cancer tissue highly expressed in the cases; PLC c-met protein expression in patients with carcinoma tissue and serum HGF concentration was positively correlated with intensity; Elevated levels of serum HGF and c-met expression in cancer tissue that probably is an important factor in the early postoperative recurrence or metastasis.
