| Objective:To study the the effect of GnRH â…¡ on expression of vascular endothelial growth factor (VEGF)ã€apoptosis and changes of volume of ectopic endometrial tissue in endometriosis rat model.Methods:(1) The establishment of endometriosis model of SD rats, and identification.(2) The model after the success of the endometriosis model of SD rats were divided into4groups, respectively given different doses of GnRH â…¡(100ugã€150ugã€200ug)to intervent, blank control group given the same volumes of saline,4weeks after the medication,ectopic lesion volume were measured with vernier caliper, the expression of VEGF of ectopic endometrial were determinated with Vernier caliper, the rate of apoptosis of ectopic endometrial cells were determinated by TUNEL method.Results:(1) The success rate of endometriosis SD rat Model is80%.(2) Different doses (ug) of GnRH â…¡ (100ã€150ã€200) were given to model rats to intervent after4weeks,the ectopic lesion volume (mm3) respectively was (63.18±9.29ã€41.12±6.06ã€24.52±8.18), which were significantly reduced compered with the volume before treatment(8.08±18.28ã€99.04±20.60,102.45±26.98), there was a significant difference (P<0.01), and with the increasing of the dose, volume was reduced more obviously (P<0.05); the volume of control group before and after treatment were(108.91±24.38ã€104.32±20.40),there was no significant difference (P>0.05).Tip:GnRH â…¡ can significantly reduce the ectopic lesion volume of endometriosis model rats, and with the dose increasing, the size is more and more small.(3) Different doses (ug) of GnRH â…¡ (100ã€150ã€200) were given to model rats to act after4weeks, the VEGF expression of ectopic endometrial cells respectively were:(0.0826±0.0019,0.0630±0.0033,0.0443±0.0024),which was significantly reduced compered with the control group (0.0949±0.0042),there was a significant difference (P<0.05); and as the dose increasing, the expression of VEGF was decreased more obvious, there was a significant difference (P<0.05). and as the dose increasing, the decreasing of expression of VEGF is more obvious, there was a significant difference (P<0.05). Tip:GnRH â…¡ can reduce the VEGF expression of ectopic endometrial tissue in model rats with endometriosis, and with the dose increasing, the expression is more and more lower.(4) Different doses (ug) of GnRHâ…¡(100ã€150ã€200) were given to model rats to act after4weeks, the apoptosis rate (%) of ectopic endometrial cells respectively were:(10.67±4.08ã€22.12±5.16ã€31.46±6.25), which was significantly elevated compered with the control group (5.32±4.23),there was a significant difference(P<0.01); and with the dose increasing, the apoptosis rate was more elevated, there was a significant difference (P<0.05). Tip:GnRHâ…¡can promote apoptosis of ectopic endometrial tissue of model rats with endometriosis, and with the dose increasing, the apoptosis rate is more and more high.Conclution:GnRHâ…¡ plays a therapeutic role through reducing the VEGF expression of ectopic endometrial tissue, promoting apoptosis of ectopic endometrial cells, and it is does-dependent manner, which together reducing ectopic lesion volume in model rats with endometriosis at last. |
