Objective To study the mechanism of vascular endothelial growth factor(VEGF) on prevention the restenosis after percutaneous coronary intervention(PCI) by observing the effect of VEGF on the apoptosis of vascular endothelial cell and the expression of Bcl-2 mRNA and Fas mRNA of vascular endothelial cell induced by ox-LDL and TNF-α.Methods Human umbilical vein endothelial cell (HUVEC) were divided into control group, ox-LDL-treated group or TNF-α-treated group and ox-LDL+VEGF-treated group or TNF-α+VEGF-treated group, the apoptosis of HUVEC was determined by flow cytometry(FCM) and in situ terminal deoxynucleotidyl transferase (TdT)-mediated deoxyuridine triphosphate (dUTP)-biotin nick end-labeling(TUNEL), the expression of Bcl-2 mRNA and Apo-1/Fas mRNA were examined by RT-PCR.Results Compared with control group, the apoptosis cells and the expression of Apo-1/Fas mRNA were significantly increased in ox-LDL-treated group or TNF-α-treated group, but the expression of Bcl-2 mRNA was markedly decreased. Inversely, in VEGF-treated groups, the apoptosis cells and the expression of Apo-1/Fas mRNA were decreased; the expression of Bcl-2 mRNA was increased.Conclusion VEGF could inhibit the apoptosis of HUVEC induced by ox-LDL and TNF-αvia downregulate of Fas mRNA expression and upregulate of Bcl-2 mRNA expression, which offered further theoretical evidence for VEGF on prevention of restenosis after PCI.
