The Research Of The Mechanism About The Treatment For Kidney Tonifying And Blood Circulation In Treating Oligoathenoteratospermia

Objective:To detect the effects of the imbanlanced of Reactive oxygen species, mitochondria on male sterility (oligoathenoteratospermia) and the correlations of them;to discuss the mechanism of tonifying kidney and activating blood (yi jing fang) to cure male sterility (oligoathenoteratospermia) from the angles of ROS,mitochondria and the sperm apoptosis; in traditional Chinese medicine angle,to study the mechanism of male sterility and mechanism that tonifying kidney and activating blood to cure the disease.Methods:Use self-control method to study oligoathenoteratospermia before and after the treatment, observe the the effectiveness of the the method of tonifying kidney and activating blood (yi jing fang),in which Sperm early apoptosis rate,mitochondrial membrane potential (MMP), Seminal plasma superoxide dismutase(SOD),malondialdehyde (MDA),UA(Uric Acid),the tongue and the pulse etc.There are29patients in the match group,while30in the experiment group:oligospermia patients5cases,simple asthenospermia patients25cases.All of them will be given drinking broth yi jing fang accordingly.Patients are followed once1-2weeks and3months for a course of treatment.Results:1The change of sperm density:All of the five both the sperm density and the sperm count of the two experiment groups are improved to a great extent after the treatment.Sperm density increases15.78M/ml on average and sperm count25M on average.2The change of the motility and vitality:after treatment, the sperm motility a level improves by an average of15.60%,(a+b) grade increases about18.87%. Sperm motility increased averagely by21.46%.3The change of the percentage of morphologically normal sperm:the percentage of morphologically normal sperm increases about11.01%on an average.4Sperm early apoptosis change:before treatment sperm early apoptosis rate (4.26±2.79)%, while after treatment early apoptosis rate (2.86±1.47)%, reduced on average1.40%(P<0.01); Live sperm (AV-/PI-)improved by an average of5.28%(P<0.05).5The mitochondrial membrane potential (MMP) loss level of the sperm:after treatment, MMP normal sperm increased by24.64%(P<0.01), MMP lost sperm decreased by19.96%(P<0.05).6Seminal plasma SOD change:before the treatment84.24±39.25μ mol/L, while after the treatment105.57±45.52μ mol/L.By contrastafter, it improved by an average of 21.33u mol/L (P<0.01) after the treatment.7Seminal plasma MDA change:before the treatment49.49±9.03nmol/L, while after the treatment39.55±7.22nmol/L.That is to say,it improved by an average of20.36nmol/L (P<0.01).8Seminal plasma UA change:before the treatment138.2±65.04μ mol/L, while after the treatment171.40±118.30μ mol/L, That is to say,it improved by an average of33.2u mol/L (P<0.01).9Correlation Analysis:the sperm motility and the sperm activity level are significantly correlated to early apoptosis,the loss of the MMP and the level of the seminal MDA,and the level of seminal MDA take up a more important role. Sperm density has a significant correlation with early apoptosis and the loss of the MMP,and there no theaf to prove they have correlation with the level of seminal MDA.Sperm early apoptosis have significant correlation with MMP,and both of them didn’t show correlation with MDA.Conclusion:The method of tonifying kidney and activating blood(yi jing fang) can improve the sperm density,motility,morphology,significantly,and reduce the rate of early apoptosis,and recover the MMP of the sperm.There are two ways at least that the method of tonifying kidney and activating blood(yi jing fang) cure OATS:1Make the circumstances of testicle better to produce more and better sperm.2Make the prostate better to produce more antoxid and make the level of ROS lower,then improve the activity level of sperm.From the perspective of traditional Chinese medicine,Kidney deficiency and blood stasis is an important factor of male infertility through the whole pathological course and a significant stage of the process.In this case,the method of nvigorating the kidney and promoting blood,with treatment based on syndrome differentiation,improves the fecundity of the patients significantly.
