Objective: Explore the anti-inflammatory and tonifying thekidney and Yang of mango nucleoli extract, and the effect of screeningthe effective parts part, determine the total polyphenol content in nucleolimango extract and effective parts, explore relationship between the totalpolyphenol and anti-inflammatory with tonifying the kidney and Yang.Methods: To use the system solvent method to separate material ofmango nucleoli extracts, divided into four parts of petroleum ether parts,ethyl acetate parts, l-butanol parts and water part. Ethyl acetate partsadopt polyamide column chromatography, gradient elution by water andethanol （from10%ethanol to90%ethanol）, to test by ferric chloride andgelatin, collecting10%flow points and50%flow points. To study theacute toxicity, anti-inflammatory, tonifying the kidney and Yang ofethanol extract and ethyl acetate parts, and to study anti-inflammatoryand tonifying the kidney and Yang of10%and50%flow points fromethyl acetate parts after the polyamide. Determinate the total polyphenolcontent by the Folin-ciocalteu colorimetric in nucleoli mango extract,ethyl acetate parts and10%with50%of flow points, considerateinfluence of these factors of the dosage of folin reagent solution, sodiumcarbonate, as well as coloration time and coloration temperature on thetotal polyphenol content, determined the best measuring conditions.Results: Mango nucleoli extract, ethanol extracts and ethyl acetate extract has some of the anti-inflammatory effects, ethanol extracts,10%flow points and50%flow points have to resist fatigue and hardy role,ethanol extracts can promote the mice’s ability to bear children, increasesperm quantity and sperm activity, can reduce the rate of sperm deformity,organ in mice increased the weight of testis+epididymis, samenblase+prostate. The standard of gallic acid and total polyphenols in760nmwavelength place has the maximum absorption wavelength. Gallic acid inthe concentration is inside of0².4μg.mL^-1, with absorbency had goodlinear relationship, in this range inside for equations of linear correlationy=0.1141x+0.0148, correlation coefficient is0.9995. The accuracy testof methods RSD value is0.604%, the RSD value of repeatability tests is1.25%, the RSD inside120min is3.60%, shows that the instrument isgood of repeatability, precision, stability. Total polyphenols of Mangonucleoli with the best conditions of Folin-Ciocalteu colorimetric detectionfor developer use level1.5mL, the dosage of10%of sodium carbonatesolution for3mL, coloration temperature30℃, coloration r time90min.Conclusion: Mango nucleoli have function of anti-inflammatory andtonifying the kidney and Yang, the effective parts of ethyl acetate parts.The relevant between the anti-inflammatory and tonifying the kidneyfunction in Mango nucleoli and its effective parts and within the totalpolyphenols, but what is the specific phenolic compounds are still needsfurther research...
