| Purpose:Rheumatoid arthritis is a kind of chronic systemic inflammatory disease. The main clinical manifestations is the chronic, symmetry, multi-synovial arthritis and extra-articular disease.It is an autoimmune inflammatory disease. In Chinese Medicine, it is called "Bi syndrome ". The manifestations are peripheral joint persistent synovial inflammation, synovial organization hyperplasia, inflammatory cells invasion, pannus formation, thereby cause the cartilage and bone destruction, cause joint deformities and loss of function, eventually lead to different degree of disability. This disease can be came on any age, the incidence is extremely broad and the progression is persistent, recurrent. This disease is a seriously threaten patients life. Its etiology and pathogenesis are not clear, so it is hard to give early diagnosis and early treatment. What’s more, the western medicine has lots of limitations in the treatment of RA, so that the curative effect is not good. However Chinese Medicine has a long experience in RA. According to many researches, Traditional Chinese Medicine has lots of merits in this area, such as in anti-inflammation analgesia, circulation improvement, immunosuppression, immunoregulationand minimal side effect.Through this project we find out the effect on hypoxia-inducible factor and vascular endothelial growth factor by the combination therapy of Traditional Chinese Medicine and Lef lunomide.. Analyse and compaire the Chinese and Western medicine therapy. The purpose is to optimize the treatment protocols and to explore a methord for the clinical therapy.Method:Divided these48rats into6groups. Build up the CIA rats models. Gavage these rats with different medicines.(trade name:Arava, lOmg/film, Suzhou Long March-Xin Kai Pharmaceuticals, Inc, Zhunzi: H20000550)2mg/day; Treatment group:Western medicine combined with traditional Chinese medicine group. On the basis medicine of control group,we added traditional Chinese medicine which are prescribed by us (the formulation are provided by Traditional Chinese Medicine Hospital of Wuhan. Soluble Granules, which are made by TCM Hospital of Wuhan itself.), melting them into water when needed,2ml/d.Dynamic observation of the following indexes:arthritis index, toe volume. Take out the rats’ankles at the36th day and conduct the HE staining and the HIF-1VEGF immunohistochemical staining. Analysis and compare The HIF-1α、VEGF changes in the synovial blood vessels of rats’ ankles.1. Arthritis index:LEF of arthritis of CIA rats havesignificant therapeutic effect, can reduce the index of ankle arthritis in CIA rats; Chinese medicine combined with LEF in the treatment of CIA rat ankle arthritis index reduce the effect more obvious.2.Joint histopathology:LEF can delay the CIA rat ankle joint synovial tissue pathological changes to reduce the generation of HIF-a and VEGF in the CIA rats in synovial tissue.
