Emilia sonchifolia (Linn.) DC. is a compositae’s annual or biennial herb, which distributes in southern of China such as Guangdong, Guangxi, Fujian, Guizhou, Jiangxi provinces. It is mild-natured, taste bitter and tiny pungent, and can be used for heat-clearing, detoxicating and detumescence. Many of flavonoids possess the activities of anti-inflammation, antioxidation, antibacterial, analgesic, enhance immunity, antiviral. Also the research proved that the flavonoids which Emilia sonchifolia contain show good performances on these aspects.In this paper, the aqueous extract flavonoids from Emilia sonchifolia are taken as the research object, and separation and purification performance of macroporous resins adsorption were investigated. The process conditions effecting on the results of purification were researched, the best conditions for the major influence factors were obtained. This research had been divided into three parts, the main content and results are given as follows:1. Hot water extracting process conditions of Emilia sonchifolia had been studied. Four factors (Extraction temperature, extraction time, extraction times, and liquid-solid ratio) were selected to proceeded the single factor experiment, and then the optimized process conditions were obtained:Extraction temperature is100℃, extraction time is3h, the extraction times is3times, the liquid-solid ratio is10:1. Furthermore, orthogonal test of4factors at3different levels were taked to optimize the process conditions with the extrating rate as the index, the result showed that when extraction temperature is100℃, extraction time is2h, the extraction times is4times, the liquid-solid ratio is10:1, the rate of flavonoids extrating is3.54%, the flavonoids content of extraction is6.05%. And the extracting temperature is greatest impact on extracting rate.2. Static adsorption and desorption performance of ten kinds of macroporous resins for Emilia sonchifolia aqueous extract flavonoids had been researched. The result showed that the HPD722macroporous resin had the best adsorption and desorption performance than others. The adsorption process of HPD722macroporous resin is exothermic, and it’s adsorption capacity with the rise of temperature and reduce, but the influence of the temperature on the adsorption and desorption is not significant. The process of HPD722macroporous resin adsorb Emilia sonchifolia flavonoids is mainly governed by liquid membrane diffusion. The first step, adsorption take place in macroporous resin surface and the large size hole’s surface, the absorption complete in a short time; the second step, adsorption continue to take place on the small hole’s surface slowly.3. The dynamic adsorption and purification peformance of HPD722macroporous resin for Emilia sonchifolia aqueous extract flavonoids indicate that the optimum conditions of purification would be:the packing glass column to the ratio of diameter to height is1:10, the quality concentration of sample solution is50.00mg·mL-1, the feeding flow rate is3.00mL·min-1, the sample pH value is4.7(original sample pH value). Absorbed with this optimum conditions, the absorbtion rate up to81.11%. And than used distilled water(the same volume with the feeding sample solution) to remove vestigial extraneous component which remain in the resins column. After that the60%ethanol(the same volume with the feeding sample solution) was used as strippant, when the eluting rate is3.00mL·min-1, it’s desorption rate reach85.26%. The content of flaoniods increased to32.65%, and the recovery of flavonoids can reach55.22%, under the HPD722macroporous resin optimum conditions of purification.
