Eupatorium odoratum is a kind of perennial herb which belong to Eupatorium genus. The whole plant can be used as traditional Chinese medicine which has many functions such as loose silt swelling, detoxification and bleeding insecticidal. The contents of flavonoids in Eupatorium odoratum were particularly rich which related to some pharmacological effects. This thesis studied on the extracting and purifying technology of flavonoids from Eupatorium odoratum, discussing the effect of flavonoids extracted from Eupatorium odoratum on antioxidation.The main content and results are as follows:Based on single experiments, extraction temperature, extraction time, ratio of liquid to solid and ethanol concentration were selected for Box-Behnken central composite designing. RSM was employed to study the effect of these factors on the yield of total flavonoids. Canonical analysis of surface responses revealed that the stationary surface was a maximum. The coefficient of determination was0.9722. The interactions among the above parameters had a significant effect on total flavonoids yield. Ethanol concentration had the most significant effect, ratio of liquid to solid took second place and extraction temperature and extraction time had a little effect. The results indicated that the optimal process conditions were extraction temperature80℃, extraction time4.5hours, ratio of liquid to solid1:41and ethanol concentration51%.Under these optimized conditions, the extraction yield of total flavonoids was up to7.162%.Results show that the AB-8microporous adsorbent resin was the best type to separate flavonoids from Eupatorium odoratum. Its static adsorption followed the Langmuir curve. The best technology of purification using AB-8microporous resin is as follows:the adsorption concentration of flavonoids from Eupatorium odoratum16%, pH5.0,1-times the column volume of80%ethanol solution to elute, expend2BV of eluting solution. After the purification, the content of flavonoids in Eupatorium odoratum will be42.34%.Flavonoids from Eupatorium odoratum had a good scavenging effect on hydroxyl radical and surperoxide anion radical, showing good dose-effect relationship at a certain concentration range. It was shown that with the increase of total flavonoids, the upward trend of POV and acid value was slow down. Flavonoids from Eupatorium odoratum can improve SOD, GSH-Px activity and reduce MDA content in serum liver and brain of mice.
