The Changes Of Myocardial Injury Markers And Inflammatory Cytokine And The Intervention Of Xuebijing Injection In Septic Rats

Objective To observe the changes of the myocardial injury markers TnT, BNP, H-FABP, CK-MB and TNF alpha of septic rats and the effects of xuebijing injection on the myocardial injury markers and TNF alpha through the animal experiment.To discuss the mechanism of myocardial depression of septic rats and the myocardial depression was improved by xuebijing injection.Methods1.Seventy eight male wistar rats were divided into four groups randomly:normal group, sham-operation group, CLP group and xuebijing-treated group.2.Blood samples of normal group were obtained from abdominal aorta after anesthesia. Blood samples of sham-operation group,CLP group and xuebijing-treated group were obtained from abdominal aorta after6,24,48and72hours of operation.The blood serum was saved after centrifugation.3.The levels of TnT, BNP, H-FABP, CK-MB and TNF-a were determined by ELISA.4.The hearts of all rats were took out rapidly after death, the ventricle muscle were cut off rapidly and were dyed by HE, observed the pathology changes of myocardial tissue by light-microscopy and photography.5. All datas were analysed by spss17.0statistical package. All datas were expressed by the mean and standard deviation. More samples compared by one-way analysis of variance, the difference was statistically significant when P<0.05. Two variables related analysis by linear correlation analysis, the relevant degree was showed by Pearson Product-moment correlation coefficient.Results1. Septic rats appeared to changes of general state, the mortality in Xuebijing group was lower than that of CLP group, but there is no difference betweem the two groups at four time points(P>0.05).2.Campared with normal group and sham-operation group,myocardial cells in the CLP group were arranged disorder, swelling,empty bubble degeneration,myocardial cell shrinks,inflammatory cells infiltration,myocardial fiber breakage and nuclear membrane rupture,the nucleus swelling,along with the time the changes were obviously,nuclear solid shrinkage, myocardial horizontal grain punch-drunk, inflammatory cells infiltrating apparently; In xuebijing group, myocardial cells are arranged owe tidy,edema and steatosis more weakened significantly than CLP group,myocardial horizontal grain is clear, inflammatory cells infiltrating fewer than CLP group.3.After CLP6h,the TnT, H-FABP,CK-MB level were increased significantly compared with normal group and sham-operation group(P<0.01), along with the time,the TnT,H-FABP,CK-MB level in serum was decreased, xuebijing injection can reduce the TnT,H-FABP,CK-MB level in serum significantly,the level was lower significantly than CLP group at different time points (P<0.01);After CLP6h, the BNP level was increased significantly compared with normal group and sham-operation group(P<0.01), along with the time, the BNP level in serum was increased, xuebijing injection can reduce the BNP level significantly, the level was lower significantly than CLP group at different time points (P<0.01), but it was higer than normal group and sham-(?)peration group.4.After CLP6h, the TNF-a level was increased significantly compared with normal group and sham-operation group(P<0.01), along with the time, the TNF-α level in serum was increased, xuebijing injection can reduce the TNF-a level in serum significantly, the level was lower significantly than CLP group at different time points (P<0.05, there was no evident difference with normal group and sham-operation group at6h and24h, but it was higer than normal group and sham-operation group at48h and72h.5.Correlation analysis shows that the level of TNF-α and TnT, BNP, H-FABP, CK-MB in rats serum were linear positive correlation obviously.Conclusion1. The septic rats model is stable and reliable which was made by cecal ligation and puncture.2.In early time,septic rats can appear myocardial injury and myocardial dysfunction obviously, performance for heart tissue pathology morphology was abnormal, the level of TnT, CK-MB, HFABP and BNP in serum was high.3. There is certain effect for TNF-α in myocardial damage.4. Xuebijing injection play a role of protection on myocardial by reducing inflammation factors level.
