Objective:To study the expression of MMP-26、TIMP-4at the villus of the normal early pregnancy and mifepristone affect the structure of vilus and the expression of MMP-26、TIMP-4. Explore the expression level and balance of MMP-26, TIMP-4in maintaining the normal pregnancy and the mechanism of mifepristone terminate pregnancy.Methods:From may2011to June2011select60noraml early pregnancy(at40-63days)women who required to terminate pregnancy at family planning outpatient of the general hospital of tianjin medical university p.Divided these women into two groups. Experimental group:give150mg points of mifepristone before Artificial abortion; control group:complete relevant inspection then Artificial abortion was performed. Observe the structure of villus throughmicroscope, immunohistochemistry methods were used to detect the distribution and expression level of MMP-26, TIMP-4in two groups.Results:1, The experimental group and control group, in age, the times of gravidity and parturition,the size of embryon there were no statistically significant (P>0.05);2, the experimental group A and experimental group B in the age, the times of gravidity and parturition there were no statistically significant (P>0.05). The size of embryon in experiment group B and group A, the difference was statistically significant (P<0.05);3, Comparison thecontrol group A and B in the age, the times of gravidity and parturition there were no statistically significant (P>0.05). The size of embryon, the difference was statistically significant (P<0.05). The time of pregnancy in control B group was longer than control A group;4, the nap trophoblastic cells degeneration and necrosis, fall off, the interstitial edema, blood deposition, cellulose deposition; The control group down size uniform, trophoblastic cellular structure is clear, the outer nourish cells for fit, the cytoplasm is rich, visible small and hyperchromatic nuclei; For cells lining nourish cells, a polygon or ovoid, the cell boundaries clear, arrangement and comparison, the cytoplasm is rich but cytoplasmic dyeing is shallow and have little bubbly of the nucleus.5, the experimental group and control group fluffy organization under mirror accidentally see sinus cavity gap that fluffy leaves of blood vessels, some lumen structure distribution in the nap and outstanding on edge lobules trophoblastic namely vascular membrane (vasculo-syncytial membrance, VSM). Both of them mainly existed in the cytoplasm of trophocyte;7, Through quantitative analysis,the expression of MMP-26in the villus of the control group was higher than that of the experimental group and the differences were significant(P<0.05);The expression of TIMP-4in experimental group was significantly higher than the control group. Difference was statistically significant (P<0.05);8, the experimental group A and B, the control group A and B MMP-26, TIMP-4expression level did not see clearly different, the difference was not statistically significant (P>0.05);9, The number of blood vessels VSM with the level of MMP-26, TIMP-4carry spearman rank correlation,relevant coefficients were r=0.027, P>0.05; R=0.037, P>0.05. So VSM number and the MMP-26, TIMP-4expression level does not exist correlation.Conclusion:1, The control group fluffy biopsy, after HE dyed in light-microscopy visible fluffy nourish cells, fluff interstitial, stroma vascular structure is clear. The role in the microsopic structure after get dramatically changed, the organization degeneration necrosis, interstitial edema.2, Both groups accidentally see fluff vascular membrane (VSM) outstanding on trophoblastic, explain to get down has no significant effect on blood essel growth.3,MMP-26and TIMP-4were expressed in villus of normal early pregnancy women, Both of them mainly existed in the cytoplasm of trophocyte. Mifepristone can influence the expression of MMP-26and TIMP-4in human villus of early pregnancy, explain the balance of MMP-26and TIMP-4play an important role for sustaining the normal early pregnancy.4,MMP-26expression decreased and TIMP-1expression increased. Get through Mifepristone break the balance between MMP-26and TIMP-4,interfere trophocyte ’s infiltrating ability and embryo implantation and placentation.Finally abortion happened.
