Objective:To explore the effect of "Xiao Shuan Ke Li"(XSKL) and also the mechanism on neonatal rats with hypoxic-ischemic encephalopathy (HIE), in order to provide the experimental evidences for Traditional Chinese Medicine in treating neonate HIE.Methods:36seven-day-old Sprague Dawley (SD) neonatal rats were randomly divided into three groups:sham operation group (n=12), observation group (n=12), model group (n=12). Animal model of HIE was induced according to the Rice method:under the microscopic magnification, the left common carotid artery was doubly ligated with5-0silk, and then sheared, after that all rats were laid back to their mothers for2h recovery.Then the rats were placed into hypoxia chamber in the environment of37℃for2.5h, with a continuous flow of8%O2+92%N2delivered at2.5L/min. While the sham operation group were only dissociated of the left common carotid without ligation or exposed to hypoxia.The rats of observation group were given oral XSKL (8g/kg, qd) for14days. Model group and sham operation group were given the same amounts of saline for14days(1) The pathomorphology changes in the left brain were observed under the light microscope, and the quantitative pathological score of three groups were compared.(2) Their expressions of Caspase-3、Bcl-2and Bax apoptosis protein in left brain were measured by immunohistoehemical staining method.Results:(1) Degeneration and necrosis were detected significantly in the left brain in the model group, less in observation group and rarely in sham operation group. The pathological score in sham operation group (22.58±2.61) was lower than observation group (32.17±1.40) and model group (50.67±2.27), each other showed significantly differences(all P<0.01).(2) Immunohistoehemical staining method results showed that the amounts of masculine cells of Caspase-3in the left brain were few in sham operation group (3.63±0.74), the masculine cells of Caspase-3in observation group (27.63±4.47) were much fewer than those of model group (33.25±4.73),each other showed significantly differences(akk P<0.01), the masculine cells of Bcl-2in the left brain were few in sham operation group(10.52±2.70), the masculine cells of Bcl-2in observation group(41.38±2.09) were much more than those of model group (22.63±3.17) each other showed significantly differences(all P<0.01), the masculine cells of Bax in the left brain were few in sham operation group(22.15±9.73), the masculine cells of Bax in observation group(49.63±18.52) were much fewer than those of model group(56.71±28.04),each other showed significantly differences(all P<0.01).Conclusions:XSKL can reduce the brain damage of HIE neotatal rats, the mechanism might be that XSKL can up-regulation of Bcl-2protein and down-regulation of Caspase-3and Bax protein. Objective:To explore the therapeutic efficacy of Xiao Shuan Ke Li(XSKL) on neonatal hypoxic-ischemic encephalopathy (HIE),by which to find new ideas and effective prescriptions of HIE.Methods:Nov.2010to Jan.2012,76neonates with HIE in neonatal ambulance sub-center were randomly divided into observation group (n=36) and control group (n=40) according to the treatment sequence. Two groups were all treated by regular therapy. In addition, neonates of observation group were added with XSKL (0.6g/kg.d,1/8h,10d) on the fourth day after birth.(1) NSE was examined at the age of4d and14d.And the changes were compared pre-treatment and post-treatment.(2) S100B was examined at the age of4d and14d.And the changes were compared pre-treatment and post-treatment.Results:(1) The clinical observation showed that the efficiency in observation group97.22%(35/36) is significantly higher than that in control group67.50%(27/40)(P<0.05).(2) Before treatment, the level of NSE in observation group(17.82±4.78)μg/L and control group(17.45±5.09)μg/L was no significantly differences (P>0.05). While the level in observation group(7.97±1.47)μg/L was significantly lower than control group (11.88±2.96)μg/L at the age of14d(P<0.01).(3) Before the treatment, the level of S100B in observation group (0.47±0.14)μg/L and control group(0.49±0.19)μg/L was no significantly differences (P>0.05). While the level in observation group (0.14±0.02)μg/L was significantly lower than control group (0.24±0.06)μg/L at the age of14d(P<0.01).Conclusion:XSKL can reduce the brain damage of neonates with HIE, to improve the clinical symptoms and reduce the level of serum NSE, S100B.
