Rhubarb(Radix et Rhizoma Rhein) which is one of the four mostfrequently used herbs, is widely used in traditional Chinese medicine, and isapplied in more than800kinds Chinese materia medica preparations. Rhubarbis widely used in clinic. Emodin, Rhein, chrysophanol, aloe-emodin andphyscion are its essential active components. Although the traditionalmedicine did not list Rhubarb as a toxic drug, reports on its adverse reactionshave been common in recent years. Abusing and using medication for too longis one of the various key reasons for an toward effects. Rhubarb contained inslimming pills and drug detoxification is major factors to long term andmisuse of the medicine. It is also reported that Rhubarb is deleterious to thereproductive system. In order to explore the rhubarb reproductive toxicity, therelationship between its toxic level and the dosage and medication time andinfluence from physiological development stage of the selected animals,based on adult rats and young rats for observation object, by testicular index,sperm counts, sperm activity rate and aberration rate for index, observedifferent doses of rhubarb water extract, total anthraquinone for different agerats reproductive functions influence. And with serum erythropoietin,testosterone, corpus luteum follicle-stimulating hormone and testis Bcl-2, Baxprotein expression level as the index, the author explores the toxic reactionmechanism of rhubarb. Designed for clinical safety, reasonable rhubarb isapplied to provide the evidence.Part1Research on correlation between rhubarb’s toxicity and dose onadult male ratsObjective: To investigate the relationship between effect and dosage of long-termRheum extracts and total anthraquinone on reproductive function of adultmale rats.Methods:Fifty SD rats with age of eight weeks were randomly divided into normalcontrol group, the0.3g·kg-1,0.6g·kg-1and1.2g·kg-1dose of water extract ofRheum palmatum group and total0.1g·kg-1of anthraquinone group. Eachcontained ten rats. The rats were given rheum extracts by gastric perfusion forthirty consecutive days, before taking out testes and epididymis to calculateviscera index and to detect sperm count, the rate of sperm mobility andaberration by hematoxylin and eosin stain and intravital staining with eosin.Meanwhile, the levels of T, LH, and FSH in serum and the expressions ofBcl-2and Bax in testes were measured by ELISA and immunohistochemistryS-P method, respectively.Results:Number and mobility rate of sperms from group RWE and RTA areobviously lower than that of the normal control group. The level of T and theexpression of Bcl-2were lower(P<0.05), and the levels of LH,FSH and theexpression of Bax were upper(P<0.05), the difference of each two group havestatistical significance.The rate of aberration was not found(P>0.05).Conclusion:Using long-term Rheum has toxic reaction to adult male rat testes, induceapoptosis, in which mesenchymal cell apoptosis significantly, resulting inreduced the synthesis of T. The tolerance of minor rats is obviously lower than adult rats. Theresearch has shown that the toxicity of rheum is more serious in developmentperiod of male animals. It has shown significant dose-dependent effect.Part3Research on the correlation between rhubarb’s toxicity andadministration days on adult male ratsObjective:To investigate the relationship between effect and day of long-termRheum water extracts on reproductive function of male rats.Methods: sixty SD rats with age of eight weeks were randomly divided into normalcontrol group, administration20days group,30days group,40days group,50days group and60days group of water extract of Rheum palmatum group,each group contained ten rats. The rats were given rheum extracts by gastricperfusion for thirty consecutive days, before taking out testes and epididymisto calculate viscera index and to detect sperm count, the rate of sperm mobilityand aberration by hematoxylin and eosin stain and intravital staining witheosin. Meanwhile, the levels of T, LH, and FSH in serum and the expressionsof Bcl-2and Bax in testes were measured by ELISA andimmunohistochemistry S-P method, respectively.Results:Compared with control group, weight and index of testis of40days,50days and60days group were lower(P<0.05). The sperm count, the rate ofsperm mobility, the level of T and the expression of Bcl-2were lower(P<0.05), and the levels of LH, FSH and the expression of Bax were upper(P<0.05), the difference of each two group have statistical significance. Therate of aberration was not found (P>0.05). The difference between50daysand60days group is not statistically significant (P>0.05).Conclusion:Rheum of different dosing days has toxic reaction to adult male rat testes,and has shown significant dose-dependent effect.
