Objective:To observe the expression of osteopontin (OPN) and Nuclear Factor-κB (NF-κB) by making rat model of abdominal aortic aneurysm and explore the molecular mechanism of abdominal aortic aneurysm.Methods:Rat model of abdominal aortic aneurysm was made by perfusing Pancreatopeptidase E from hog pancreas into the aorta of rats. Aortas were taken out two weeks after perfusion. To measure the diameter of all aortas, we used a needle as a standard of comparison. Numbers were then obtained by proportion of the aorta and the needle in photos we took and the actual width of the needle. HE staining were used to observe the structural change and Victoria staining to observe how the area of elastase altered in the middle tuna of the aorta. Immunohistochemistry and western blot were used to detect the expression of OPN, NF-κB and MMP-2.Results:Rat model of abdominal aortic aneurysm was successfully made. HE staining and Victoria staining showed that comparing to saline group and normal group, the infiltration of inflammatory cell was more apparent and the quantity of elastase was significantly lower in experimental group; Immunohistochemistry results showed that the expressions of OPN were7.5×1012±2.92×1011、3.07×105±2.58×105、0(F=6.59, P=0.004); The expressions of NF-κB were2.36×1010±2.80×1010、0、0(F=6.96, P=0.004) and the expression of MMP-2weres4.52×1010±1.73×1010、0、0(F=6.83, P=0.003); Western Blot showed the expressions of OPN were2.77×105±2.82×105、4.52×103±1.36×103、2.11×103±1.99×103(F=9.49,P=0.001); The expressions of NF-κB were6.014×104±6.27×104、1.34×103±1.89×103、4.35×102±7.01×102(F=8.91, P=0.001) and the expressions of MMP-2were9.25×104±6.87×104、8.00×102±7.89×102、2.70×102±3.89×102(F=17.91,P<0.001)。Conclusions:OPN and NF-κB were correlated with abdominal aortic aneurysm in rats. OPN may increase the expression of MMP through the NF-κB pathway in rat model of abdominal aortic aneurysm. Thereby accelerating the degradation of extracellular matrix and eventually lead to occurrence of aortic aneurysm.
