Background:Nerve growth factor (NGF) is first discovered target-derived neurotrophic factor,it participate in the occurrence and development of peripheral inflammatory pain and is one of the important mediators of the pain of peripheral sensitization Adenomyosis is an estrogen-dependent diseases..Studies have shown that estrogen stimulate the cyclooxygenase-2(cox-2) to PGE2increased, leading to pain. COX-2is a membrane structural proteins which is rapidly synthesized only when cells are stimulated, not resting expression.Vascular endothelial growth factor (VEGF) and is the most important to promote one of the angiogenic factors occur in the EM and AM playing an important role in the development. Our group preliminary study found that NGF induced dysmenorrhea in adenomyosis, and the results suggest that the lesions local expression of NGF in the intensity and the severity of dysmenorrhea is a positive correlation. COX-2or VEGF whether induced dysmenorrhea? This subject will study the expression of NGF, COX-2and VEGF to further explore the mechanism of NGF-induced dysmenorrhea. Objective:The objective of this subject is by analyzing the local lesions with adenomyosis of NGF and COX-2and VEGF and the expression of serum and NGF IL-6TXB-2,6-Keto-PGF1α to understanding the correlation of NGF and COX-2and VEGF in the mechanisms of dysmenorrheal, to further explore the mechanism of NGF involved in dysmenorrhea.Methods:1)the age is between25and50years old, Three months prior to surgery did not use any hormone therapy and not associated with other endocrine diseases, or immune system diseases2) Adenomyosis group:25hysterectomy patients because of dysmenorrheal be VAS dysmenorrhea scores before surgery, and whichever the serum5ml ELISA detection of NGF, IL-6TXB-2,6-Keto-PGFlα expression. Taken adenomyosis lesions by immunohistochemical method in the intraoperative detection of NGF, the expression of COX-2and VEGF3) in the control group:25hysterectomy patients because of Uterine fibroids or CINIII be VAS dysmenorrhea scores before surgery, and whichever the serum5ml ELISA detection of NGF, IL-6TXB-2,6-Keto-PGFlα expression. Taken the normal endometrium by immunohistochemical method in the intraoperative detection of NGF, the expression of COX-2and VEGF statistical methods:To adopt SPASS13.0software for statistical analysis, groups were analyzed using the χ2test, the indicators are based on the x-±s, P≤0.05as significant difference in the standard; and correlation statistical analysisResults:1)Dysmenorrhea in patients with lesions of adenomyosis of NGF and COX-2and VEGF in expression higher than that of the control group (P<0.01); The Adenomyosis lesions of NGF expression and The severity of dysmenorrhea is associated (R=0.4080P=0.0429). The expression of COX-2,VEGF and the severity of dysmenorrhea was no significant correlation.2) Adenomyosis dysmenorrhea serum NGF expression of IL-6TXB-2, higher than the control serum (P<0.05),6-Keto-PGF1α expression was significantly lower than the control group (P<0.05), and all of them was no significant correlation wth dysmenorrhea (P>0.05). Conclusions:NGF induced dysmenorrhea, the high expression of NGF、COX-2and VEGF may be involved in the occurrence and development of the adenomysis disease.Speculated that the three may be the reason of nerve and vascular abnormal which lead to the occurrence of dysmenorrhea.
