According the theory of TCM, the anti-inflammatory function of rosmarinic acid (RosA) is researched systematically from the level of entire animal, tissue and organ, cell and molecule by means of advanced modern technology such as analysis, cell culture and ELISA, and the anti-inflammatory mechanism of RosA was conducted in this paper. providing a theoretical basis when RosA was better applied to health food, anti-inflammtory drugs and cosmetic.First, inflammatory cell model was established by LPS acting on the macrophages in order to study the anti-inflammatory function in vitro. The result showed that RosA derceased markedly the content of TNF and NO in a dose-depent manner.Secondly, the anti-inflammatory function was studied on many animal models, whether the acute model characterized by swelling, emigration of WBC, capillary permeability or the chronic model characterized by hyperplasis of granulation. The result showed that RosA had a marked suppressive effects on normal mouse’s and ablated adrenal gland mouse’s ear edema induced by xylene, the mouse’s hind paw edema induced by carrageenin, the increased vascular permeability caused by acetic acid, the mouse’s granuloma caused by cotton ball. It was showed that had a marked anti-inflammatory function.Finally, the anti-inflammatory mechanism of RosA was researched tentatively. The result showed that the activity of superoxide dismutase (SOD) and catalase (CAT) in serum were enhanced by RosA, the content of NO、IL-1、IL-8and TNF in serum were decreased by RosA, and RosA significantly decreased MDA、PGE2、Histamine and5-HT, the activity of IL-1、TNF and PLA2were determined. RosA had no significant effects on the weight of pituitary and had no significant effects on the weight and level of VC of adrenal gland of mouse. These results suggested that the the anti-inflammatory mechanism of RosA was due to the synthesis or release of free radical and inflammatory mediators and cytokine, but no related to pituiyary-adrenal system.
