Millettia specisoa Champ is produced as an important medicinal plant resources in subtropical regions. Millettia specisoa Champ has a long history, according to "Lu Chuan Materia Medica":"can removing heat from th lung to relieve cough;, cool detoxification"; regarded as food supplements as well as drugs in southern China, so there "Medicinal and Edible," one said. Clinical certificate of Chinese Medicine has confirmed its role in the treatment of many chronic diseases, used for the treatment of lumbar muscle strain, rheumatoid arthritis, chronic hepatitis, weakness and so on; suggests the potential treatment of hepatitis, anti-vascular inflammation and antioxidant. From the literature review, we found that in recent years has completed a great cattle chemical analysis and studies on potential basis part of the active ingredient, but research on the pharmacological effects of the Millettia specisoa Champ only stay in vitro analysis and cellular level, but pharmacological studies in vivo has rarely been reported, the specific molecular mechanism of action remains unclear, which need further study. In this paper, the use of laboratory instruments to the functional ingredient contained in the Millettia specisoa Champ extraction has been separated. Generation models had been built, compare the differences in vivo and in vitro. And the efficacy of extraction and anti-inflammatory mechanism was to conduct a preliminary discussion. Study details and results are as follows:1. anti-inflammatory extract and determination of the active ingredientIn accordance with the chemical structure of Millettia specisoa Champ were extracted mainly ultrasound-assisted, low-temperature extraction, precipitation and recrystallization methods, and the use of dialysis, TCA and other methods were purified, isolated Millettia specisoa Champ polysaccharide (PMSC) and flavonoids (FMSC) two components. PMSC which measured the total sugar content of 92.43%, FMSC flavonoids content of 81.24%.2. The antioxidant activity of FMSC was measured in the experiment.FMSC could highly reduce the DPPH·、OH·、O2·, the value of IC50 was 31.50 μ g/mL,225.04 μ g/mL,115.39 μ g/mL, all lower than ascorbic acid. However FMSC got strong reducing power and chelating of iron. FMSC could depress the BUN (29.63%) in serum and elevate the activity of T-AOC (58.89%)(P<0.05); Elevated the activity of T-SOD (43.99%)、CAT (47.33%) and GSH-Px (40.76%) and the content of GSH (62.17%) (P<0.05), suppressed the content of MDA (36.53%) (P<0.01). The antioxidant activity of FMSC are very different in vitro and in vivo, however, the FMSC got excellent oxidant resistance, stronger in vivo and could be multifunction in antioxidant activity.3.The anti-inflammatory activity of PMSC was measured in the experimentTo determine the liver protection and anti-inflammation of polysaccharide in Millettia speciosa Champ, established acute liver injury model in mice. The result shows, significant inhibitory effect of liver injury were observed, the inhibition rate was up to 72.18%. The tissue sections also indicated protective effect of liver. The results showed a significantly reduction of ALT and AST activity(P<0.01), significantly suppressed the formation of MDA(P<0.05) and the expression of COX-2(P<0.05), depressed the synthesis of inflammatory mediator. Indicated the effect of liver protection and anti-inflammation of polysaccharide in Millettia speciosa Champ, possessed good application prospect.4. synergism of two components in Millettia specisoa ChampModel of inflammatory mediators induced by in vitro metabolism was built to study the role of PMSC and FMSC single and combined effects. The results showed that, PMSC could inhibit the PRP suspension inflammatory mediators PGE2 (57.9%) produced, this inhibition is independent of FMSC; FMSC and PMSC generate platelet-derived NO has a certain inhibitory effect, inhibition rate was (71.55%,23.11%).Anti-inflammatory of Millettia specisoa Champ is related to mechanism, effect of protects the body cells and inhibition of lipid peroxidation and COX-2/PGE2 metabolic pathways, also by enhancing the body’s antioxidant capacity, inhibiting the release of a large number of harmful ROS plays an anti-inflammatory effect.
