[Backgroud and Objective]Cervical carcinoma is the most common carcinoma among females,and take the first place in certain developing countries.As the launching of general investigation and general treatment, mortality of cervical carcioma significantly declines. However there are growing more young woman with cervical carcinoma.which is a heavy burden to domesticity.It is know,with developement of cell and molecular biology recently,as tumorigenesis and development which involves in in many tumor molecules is muti-stage and step complex process.Caveolae gene is a tumor supressor gene and can encode caeolin-1protein.it is a scaffolding protein in cytomembrane that is relevant with tumor cell proliferation, conglutination,metastasis and apotosis. It was found that the missing or frature of caveolae gene in tumor such as hepatic cancer, ovarian cancer,breast cancer.gastric carcinoma,et c promt that caeolin-1is relevant with tumorigenesis and development. Caveolin-1experssion is low in ovarian cancer.and the worse differentiation and the latter clinical stage is,the lower expression is.In prostatic cancer, caveolin-1as a tumor maker can be used in early diagnosis and prognostic value,and blazes a new way to do early detection and treatment. The expression of caveolin-1in renal cell carcinoma is relevant with survival prognosis.The higher the expression is,the worse the prognosis is;on the contrary the lower the expression is,the better the prognosis is.It is concluded from the above research that caveolin-1is conducive to the development and differentiation of cell.while in the process of transformation and proliferation, the expression level of caveolin-1is down, it may promote the tumor invasion and metastasis.There are controversy about caveolin-1expression in tumor and few literatures about expression of caveolin-1in tumor,espacially in cervical carcinoma home and abroad.VEGF is a high sepcial vascular endothelial cell mitogen.it play biological function by combination with special receptor and a series of signal transduction.Recent research found that VEGF is a important tumor angiogenesis modulating factor.which correlates with tumorigenesis,development and metastasis.Though many kinds of cell express VEGF that mainly exerts on hyperplasia state of the vascular endothelial cells.only tumor cells can release VEGF.In the development of cervical cancer,angiogenesis developement which is the premise of tumorigenesis and development plays a important role. New vessels provide a way for tumor to intake nutrient and at the same time for tumor cells metastasis.VEGF is an angiogenesis accelerator,while caveolin-1is an target point of angiogenesis accelerator and supressor. Caveolin-1plays important role in angiogenesis of VEGF/VEGFR2.However,there is no lierature about the relevance of caveolin-1and VEGF home and abroad.This experiment used immunohistochemistry study the expression of Caveolin-1and VEGF in normal cervix.cervical intraepithelial neoplasia(CIN) and cervical cancer.and furthermore analyze the relation between the expression of caveolin-1and VEGF and clinicopathology.Discuss the potential relationship of Caveolin-1and VEGF in tumorigenesis,metastasis of cervical cancer;and explore the significance of Caveolin-1and VEGF in early diagnosis and prognosis of cervical cancel.[Method]130cases of paraffin specimens were collected from our hospital between January2008and Febrary2012.All cases has complete clinicopathy data.40cases of chronic cervicitis,30cases of cervical intraepithelial neoplasia I/II(CIN Ⅰ/Ⅱ) consisted of CIN I12cases and CIN Ⅱ18cases,20cases of cervical intraepithelial neoplasia Ⅲ (CIN Ⅲ) and40cases of cervical cancel.21cases of stage Ⅰ,19cases of stage Ⅱ.sepectively.All cases,40cases of normal cervial tissue from fibroid as control group, was used immunohistochemistry study the expression of Caveolin-1and VEGF in normal cervix.cervical intraepithelial neoplasia(CIN) and cervical cancer,and furthermore analyze the relation between the expression of caveolin-1and VEGF and clinicopathology.Discuss the potential relationship of Caveolin-1and VEGF in tumorigenesis,metastasis of cervical cancer;and explore the significance of Caveolin-1and VEGF in early diagnosis and prognosis of cervical cancel.All statistical analysis,including Chi test,Fisher probabilities test, was performed using the SPSS13.0software.[Result]1、Immunohistochemistry(IHC) showed the positive chromatosis of Caveolin-1protein in cervical tissue were all located in the cellular membrane and kytoplasm and is claybank or sepia.The positive expression rates of Caveolin-1in normal cervical tissue(NCT),chronic cervicitis,CIN Ⅰ/Ⅱ, CIN Ⅲ and cervical carcinoma (CC) were100.00%(30/30)、95%(38/40)、83.33%(25/30)、75.00%(15/20) and the were sigmificant difference between the nomal72.50%(29/40), showed significant difference between the five groups (P<0.05); compared with CC group, there were significant difference between NCT group and CC group (P<0.0125), but no in others.The expression of Caveolin-1in cervical carcinoma showed significant correlation to age and the clinic stage of FIGO (P>0.05),but showed statiscal significance in tumor volume.Edmondson’s grade,invasion depth,and regional lymph nodes metastasis (P<0.05);and by further logistic regression analysis, the expression of Caveolin-1in cervical carcinoma showed significant relevance with tumor volume,invasion depth, and regional lymph nodes metastasis.The bigger tumor shows lower expression (OR=0.052, P<0.05);the deeper invasion shows lower expression (OR=0.044, P<0.05); the higher expression in regional lymph nodes metastasis than no regional lymph nodes metastasis (OR=0.037, P<0.05)2、Immunohistochemistry(IHC) showed the positive expression of VEGF protein in cervical tissue were mainly located in the kytoplasm and is claybank or sepia.The positive expression rate of VEGF in NCT.chronic cervicitis,CIN Ⅰ/Ⅱ, CIN Ⅲ and CC were10.00%(3/30)、15.00%(6/40)、26.67%(8/30)、5.00%(9/20) and70.00%(28/40), respectively,showed significant difference (P<0.05) in inter-group comparisons (P<0.05); compared with CC group, there were no significant difference between group CIN Ⅲ and CC group (P>0.0125), but in others.The expression of Caveolin-1in cervical carcinoma showed no significant correlation to age,tumor volume and the clinic stage of FIGO (P>0.05).but showed statiscal significance in,invasion depth, Edmondson’s grade and regional lymph nodes mctastasis (P<0.05); and by further logistic regression analysis, the expression of Caveolin-1in cervical carcinoma showed significant relevance with and regional lymph nodes mctastasis. The better Edmondson’s grade shows lower expression (OR=0.156,P<0.05); the higher expression in regional lymph nodes metastasis than no regional lymph nodes metastasis (OR=0.024,P<0.05) 3、The negative correlation was noted between Caveolin-1and VEGF proteins in cervical careinoma and CIN III group (P<0.05), while there is no correlation in chronic cervicitis and CIN I/II (P>0.05)[Conclusion]1、The expression of Caveolin-1protein gradually degrades expression in normal cervical tissue(NCT),chronic cervicitis,CIN I/II, CIN III and cervical carcinoma (CC).The expression of Caveolin-1protein in cervical carcinoma (CC) is significantly relevant with tumor volume, invasion depth and regional lymph nodes metastasis.The down-expression of Caveolin-1is an early event in cervical tumorigenesis and development,probably associated with the tumor metastasis.2、The expression of VEGF protein gradually degrades expression in normal cervical tissue(NCT),chronic cervicitis.CIN I/II. CIN III and cervical carcinoma (CC).The expression of Caveolin-1protein in cervical carcinoma (CC)is significantly relevant with pathological grade and regional lymph nodes metastasis.The down-expression of Caveolin-1is an early event in cervical tumorigenesis and development.probably associated with the tumor metastasis.3、The expression of Caveolin-1protein presents negative correlation to VEGF proteins in CIN III and cervical carcinoma,manifesting mutual counteractions of the tumorigenesis,development and angiogenesis.
