OBJECTIVE:In this study, the proposed intermittent upright position established the rabbit intervertebral disc degeneration animal models to explore the intermittent vertical build the feasibility of disc degeneration model and the model is simple and easy.Through this model to analysis on the degeneration of the rabbit lumbar intervertebral disc’s pathology, imaging, PG, and dynamic analysis of MMP-3, TIMP-2in nucleus pulposus and annulus fibrosus, searching for the etiology and mechanism of intervertebral disc degeneration in favour of prevention and treatment of low back pain caused by lumbar disc degeneration.METHODS:42New Zealand White rabbits, randomly divided into the experimental group (n=21) and the control groups (n=21). In the experimental group the PVC pipes were uprighted and in the control group the PVC pipes were flated, all of them are fixed5hour in everyday. In4,8,12weeks, the two groups of rabbits respectively choose7on the spine (L4/5as the center)X-ray, MRI, PG, HE observation of intervertebral disc degeneration, and in different experimental period analysis and comparison of nucleus pulposus, annulus fibrosus of MMP-3, TIMP-2change.RESULTS:In the molding process, the42rabbits without loss, were all involved in the result analysis. At4weeks, between the experimental group and the control group differences in disc height index, MRI and annulus fibrosus changes with no obvious meaning (P>0.05). At8,12weeks of the experimental group and the control group of intervertebral disc height index compared to the difference is significant, lumbar intervertebral disc degeneration in the T2weighted image signal was significantly reduced, hematoxylin-eosin staining showed annulus fibrosus derangement. In experimental modeling groups,4,8,12weeks in the nucleus pulposus tissue by PAS stain showed degenerative nucleus pulposus gray value significantly larger (P<0.05).In the annulus fibrosus, the control group in the same time the MMP3and the TIMP2average MOD value compared with no statistical significance (P>0.05); with the modeling of time to extend the time period in the experimental group the value of TIMP2than MMP3average MOD value significantly (P<0.05), in which12weeks the average MOD value differences there is significant (P<0.01).At the same time comparison of different groups, the control group in MMP3average MOD value in8weeks had no significant difference (P>0.05). In12weeks average MOD changes significantly in value than8weeks (P<0.05); the TIMP2average MOD value with the molding time changes obviously, in8weeks when there was not significant difference (P<0.05), in12weeks difference changes significantly (P<0.01).In the experimental group MMP3along with the molding time MOD value increases gradually (P<0.05), while TIMP2average MOD value in8weeks relative to4weeks had no significant difference (P>0.05), the12weeks average MOD values increased (P<0.05).In the nucleus of MMP3, TIMP2average MOD values indicate, in the control group and the experimental group at the same time MMP3and TIMP2average MOD value compared with statistical significance (P<0.05).In the same group in different time periods were compared the results show that with the extension of time, the control group MMP3and TIMP2average MOD value in8W had no statistical significance (P>0.05), the12weeks average MOD values were increased significantly (P<0.05); the experimental group MMP3and TIMP2average MOD value has significant difference (P<0.05), and MMP3in12weeks average MOD value increased significantly (P<0.01).CONCLUSION:1. By forcing the upright position due to their own body under gravity loading on the intervertebral disc to produce the effects can be successfully established model of intervertebral disc degeneration.2. Change in a variety of factors have led to the disc histology affect the endplate first, followed by the nucleus pulposus, and finally the annulus.3.MMP3mainly in the nucleus pulposus in expression, and TIMP2mainly in the fibrous rings of expression, which may and its effect on.
