Armillaria mellea (Vahl ex Fr.) Quel is a kind of famous medicinal and edible fungus belong to Tricholomataceae Armillaria(Fr.:Fr.)Staud. Armillarl’s fruiting bodies has the function of to hypnotize, calm, anti-vertigo, to improve heart and cerebral blood and to circulaationto safeguard the brain. Armillaria mellea can therapy many kinds of diseases. Armillaria mellea is a kind of Chinese crude drug which was used on nutritions and health care in our country. Especially its chemical composition was constantly scholars of home and abroad investigative emphasis. Armillaria mellea fermentation broth as the part of Armillaria mellea’s resources didn’t systemic report on study and research. Respecting this, the author deeply studied and researched the chemical composition of Armillaria mellea fermentation broth, in other to clear-cut the effect of the in force part. Our purpose is developly Chinese tradition medicine and health products in accordance with market demand. This is also a goal of modernization for Chinese tradition medicine. The following aspects were included:First of all, Researching progress of the Armillaria mellea’s chemical constituents and pharmacologic actions by literature search, which is in other to provide more information on research the chemical constituents and pharmacologic activeness of Armillaria mellea fermentation broth.Second, volatile constituents were extracted by circumfluence used diethyl ether and We have identified41chemical composition with GC-MS, which represented94%of the total GC peak area of the volatile constituents. There were Furaldehyde, Ketones, Esters and Coumarins. By this experiment we can understand the hylic substructure of Armillaria mellea fermentation broth, and get information about chemical constituents Armillaria mellea fermentation broth.Third is chemical constituents’ analysis. The extract of Armillaria mellea fermentation broth by water was divided into four fractions:Petroleum Ether, Ethyl acetate, n-butyl alcohol and water. There are four compounds in the fractions of ethyl acetate, they were to separate by Silica gel column chromatography, Sephadex column chromatography, and prepare liquid phase. Two of them were the firsi time get from Armillaria mellea fermentation broth, there are5-(hydroxymethyl)-2-Furancarboxaldehyde, and cirsiumaldehyde.Fourth, to optimize the extraction’s Industrial Arts of Armillaria mellea fermentation broth. Throw the result of single factor and orthogonal experimental design, to decide the best extraction’s Industrial Arts by the extraction ratio of5-HMF. The optimum extraction condition is that the dosage of extraction solvent is6times, two-time extraction for20min each time.Fifth, the extraction, separation, purification and structures’research of Armillaria mellea fermentation broth’s polysaccharide. To extract polysaccharides from the fermentation broth of Armillaria mellea, by water extraction and alcohol precipitation method, polysaccharide was refined by different concentrations of ethanol and alcohol precipitation classification system, through Sephadex (Sephadex G-200) chromatography to obtain homogeneous polysaccharide AMFP3-2and AMFP4-2. Assaying purity and molecular weight determination were used high performance liquid chromatography; monosaccharide composition was used pre-column derivatization HPLC method. The consequence shows AMFP3-2and AMFP4-2’s Peak position molecular weights were258929D and1828D. Homogeneous polysaccharide AMFP3-2was composed of mannose, rhamnose, galacturonic acid, glucose, galactose, xylose, arabinose. Homogeneous polysaccharide AMFP4-2was composed of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, arabinose. We can resolve the AMFP3-2’s structure by partial acid hydrolysis, methylation, ultraviolet spectrum, infrared spectrum and NMR. AMFP3-2’s principal conformation is galacturonic acid and galactose, thire connection mode is (1â†'6) and (1â†'3,6).Finally, to research the pharmacologic activeness of the micromolecule’s fractions, the micromolecule is from Armillaria mellea fermentation broth. We study the fractions of ethyl acetate which is in Armillaria mellea fermentation broth. We had demonstrated the micromolecule which is in Armillaria mellea fermentation broth has the effect of relieved pain, by the methods chemistry and heat stimulation. Also has protective effect for cerebral ischemia by to obstruct the bilateral arteries of bandicoot rats and head-breaking of mice. We had also demonstrated it has the effect of anti-anoxia by how long time the mice life in the anti-anoxia’s surroundings. The consequences show the composition of the ethyl acetate extractive, have conspicuous analgesic activity and protection cerebral ischemia activity, and also can ignificant prolongation the time fo live in the anoxic environment.
