Objective:This topic select Panax notoginseng saponins(PNS) and Panax notoginseng powder to adjust the intervention of asthmatic mice model, observe the pathological changes of asthmatic mice lung tissue and Th lymphocyte’s cytokines IL-4, IFN-y, IL-17expression changes.Explore the effect of Panax notoginseng preventing and controlling asthma. Provide new ideas and preliminary research foundation for Panax notoginseng clinical applications.Methods:SPF BALB/c female mice80,6to8weeks of age, body weight (20±2) g, the experimental animals were randomly divided into normal control group (A), asthma model group (B), high, medium and low dose groups of Panax notoginseng powder (C. D, E. group), high, medium and low dose groups of PNS (F,G,H group). The mice in the normal control group and drug intervention groups were intraperitoneally injected with0.2ml sensitized solution (contains50μg OVA)on the1st,8th and15th day, and stimulating solution with2%OVA were consecutively given by aerosol inhalation from the22nd to28th day to stimulate the asthma attack. Sensitization and stimulation were performed with physiological saline in the normal control group. During the stimulating period (the22nd to28th day), PNS and Panax notoginseng powder were intragastrically given every day to the mice in the intervention groups30minutes before the aerosol inhalation of2%OVA. Physiological saline was intragastrically given in the normal control and asthma model group.24hours later take the blood and lung tissue of mice. Observe lung tissue pathological changes. Detect IL-4and IFN-γ, IL-17concentration in blood serum by ELISA method.Results:1. Mice behavior changes during excitation process:Normal control group (A) mice showed no abnormal changes after excitation. Asthma model group (B), drug intervention group excited visible asthma-like symptoms, the more obvious in the model group and drug intervention group eased.2. Lung tissue pathological observed results:Group lungs bronchioles and perivascular had been seen a large number of inflammatory cell infiltration, structural disorder of the airways, mucus exudate increased in the tracheal cavity significantly, the alveolar septal expand, surrounding tissue edema and inflammatory cell infiltration in the asthma model. And the bronchial lung tissue and alveolar structure clear, airway mucosa epithelial integrity, and no obvious inflammatory cells invasion embellish in the normal control group.Different degrees of ease in the drug intervention groups.3. IFN-γ, IL-4, IL-17concentration change by PNS:PNS with low and medium dose group can increase cytokines IFN-y expression, compared with the asthma model group, the difference was statistically significant (P <0.05,P<0.01). PNS high dose and medium dose can reduce the concentration of IL-4, compared with the asthma model group, the difference was statistically significant (P<0.01. P<0.01). PNS medium dose can reduce the concentration of IL-17, compared with the asthma model group, the difference was statistically significant (P<0.05).4. IFN-γ, IL-4, IL-17concentration change by Panax notoginseng powder:Panax notoginseng powder medium dose can increase cytokines IFN-y expression,,compared with the model group, the difference was statistically significant (P<0.05). Panax notoginseng powder low, medium and high dose can reduce the concentration of IL-4, compared with the asthma model group, the difference was statistically significant (P<0.01, P<0.01, P<0.01). Panax notoginseng powder each dose group were to reduce the concentrations of IL-17. But the difference was not statistically significant (P>0.05) compared with the model group.Conclusions:1. The experiments proved that PNS and Panax notoginseng powder can lessen the lung tissue of asthma mice bronchi and perivascular inflammatory cells infiltration and reduce airway epithelial mucus secretion, improve the state of airway inflammation.2. One of the mechanism of PNS and Panax notoginseng powder control asthma is may be through increase the secretion of IFN-γ, inhibit the secretion of IL-4.Then promote the differentiation of ThO to Th1, restrain the differentiation of ThO to Th2. make balance state of Th1/Th2recovery.3. PNS and Panax notoginseng powder may be through inhibit the expression of IL-17to reduce airway inflammation, intervention asthma attack.
