Purpose: Observe the various index variation of nonalcoholic fattyinflammatory, fibrosis damage and the endoplasmic reticulum stress,which are formed under high-fat diet, the paper also discussed theprotective mechanisms of DHA on liver cells.Methods:the study from two aspects,on the one hand,take SD malerat as the research object,and respectively set four groups:negative blankgroup, the positive high fat group, DHA high-dose intervention group,DHA small doses intervention group.Make four weeks and eight weeks,feeding respectively, as observation time points, through the analysis ofthe liver form, HE dyeing,oil red O dyeing,Masson dyeing,biochemicalindex test, liver enzymes test and Real-time PCR method to observe andanalyse the damaging effect on liver cells of a high-fat diet and DHA as theprotective mechanism. On the other hand, take L02cells as the researchobject,by cell microscopy,oil red O dyeing and the test of liver enzymesto determine the saturated fats’ incubative concentration in the model ofhigh fat cells. Then through the gradient analyses of the saturated fattyacid’s incubative time, finally set3groups,including: the positive highfat group, DHA intervention group, PBA intervention group, Use themethods, such as Real-time PCR method,to observe the changes of eachgroup’s all kinds of factors:Results:In animal experiments,compared with negative blank group,the positive high fat group’s lipid largely gathered, inflammatorycontaminated appear in large Numbers,and there is part of the fibrosis,biochemical index also shows the total triglycerides,total cholesterol andlow density lipoprotein cholesterol increases, high-density lipoproteincholesterol decreases,and this group’s liver enzymes generally increased,inflammatory index factor, fibrosis indices factor and UPR index factorgenerally rised. DHA intervention group, especially DHA small dosesintervention group, in lipid gathered, cells form, biochemical index,liver enzymes and inflammatory index factor,fibrosis indices factor andUPR index factor shows a better intervention role compared with positivehigh fat group. In the cell experiments, in high fat positive group andpositive control group, each inflammatory index factor, fibrosis indices factor and UPR index factor generally increased,DHA intervention group,PBA intervention group and various factors universally decreased,Conclusion:Through the two aspects of the experimental analyses,itis sure that high-fat diets lead liver cell lipid drops to gathered themselvestogether, and change the composition and structure of the original lipidcells,thus constantly stimulating UPR and lead to ERS,eventually causedthe nonalcoholic fatty hepatitis, fibrosis damage, whereas the DHA,through changing the structure of fatty acid in diet reduces lipid’s gatheringto liver cell and then reduce UPR level in order to delay the development ofthe nonalcoholic fatty hepatitis and fibrosis....
