The Effect For Nourishing Yin And Supplementing Yang Decoction Method To The Expression Of GDF-9mRNA And BMPR Ⅱ MRNA In The PCOS Rat And Medicated Serum To The Culture Of The Follicle Of PCOS Rat

Polycystic ovary syndrome(PCOS) is a common disorder in reproductive function for women, affecting approximately5%-10%of women during their childbearing years.It is the main reason for ovulation inhibition, infertility, irregular menstruation for women. Because of its underlying etiology remains unknown and the pathophysiology is unclear, also lack of more effective treatments, people from modern medicine think that it should deep into local ovarian endocrine factors, immunology, genetics, so it is received increasingly more attention for Medicine scholars.Hyperandrogenism, Hyperluteinizing hormone, hyperinsulinemia and insulin resistance is the most important endocrine characteristics in PCOS. The causes of these characteristics are too intricacy, study shows that follicular development is not only the material basis of fertility, but also the key to the crux in women with PCOS. Because of its complicacy and clinical manifestation of polymorphism, it makes this treatment more difficult. The study shows Chinese medicine treatment have some advantages and great potential.Since the present understanding for PCOS animal model, establishing PCOS rat model, on the one hand through the way to mechanically dissect and culture preantral follicle; on the other hand through pouring Nourishing yin and Supplementing yang Decoction into stomach to treat PCOS, detecting the difference in expression of GDF-9mRNA, BMPRⅡ mRNA and serum hormone in the PCOS rat, thus we can clarify its action from molecular mechanisms.The result shows that level of testosterone in group B is significantly higher than in group A, after administration, as compared to group B, the level in groupC, ZH,BH is significantly lower; the level of LH in group B is higher than in group A significantly, after administration, other groups except group BL and BM are decreasing significantly; the level of FSH is lower than in group A significantly, after administration, all treatment groups become increased significantly. The expression of GDF-9mRNA is lower than that in group A significantly, after administration it becomes to appear different degree of rising; the expression of BMPR Ⅱ mRNA is higher than that in group A significantly, after administration it becomes to appear different degree of decreasing. Using medicated serum to cultivate preantral follicle in PCOS ovary, we found that other groups except group A became different degree of obstacles after two days, the granulose cell layer reduced, the follicular development in group BH was better than other groups, it indicated the high-dose of Supplementing yang Decoction may promote follicular development.
