Objective:In recent years, the incidence of the cervical cancer is rising gradually and tendsto be younger. This article aims to study the expression of CD44v6and Ezrin innormal cervical tissue, cervical intraepithelial neoplasia organizations and cervicalsquamous cell carcinoma tissue, and detect the respective express the relationshipbetween the level of cervical squamous cell carcinoma clinicopathological featuresrespectively, then explore their roles in the development, invasion and metastasis ofcervical squamous cell carcinoma, providing a theoretical basis for the treatment anddetermining tumor malignancy and prognosis of cervical squamous cell carcinoma,supplying a new target for the further treatment of cervical squamous cell carcinomaas well.Methods:Randomly selected the cervical tissue specimens from the outpatient andinpatient in China-Japan Union Hospital of Jilin University during2010January to2012October. Making HE staining, the CD44v6immune staining and Ezrinimmunohistochemical staining for all samples. To detect the expression andrelationship of CD44v6and Ezrin in normal cervical tissue, cervical intraepithelialneoplasia organizations and cervical squamous cell carcinoma to do statisticalanalysis.Results:1、The positive expression rate of CD44v6in normal cervical tissue, cervicalintraepithelial neoplasia and cervical squamous cell carcinoma was33.33%,45.83% and68.97%, respectively. There was no significant difference of CD44v6betweennormal cervical tissue and cervical intraepithelial neoplasia organization (x2=0.667,P>0.05). There was statistically significant difference in expression of CD44v6between normal cervical tissue, cervical intraepithelial neoplasia organizations andcervical squamous cell carcinoma (x2=7.300, x2=3.863, P<0.05), it was statisticallysignificant.2、The positive expression rate of Ezrin in normal cervical tissue, cervicalintraepithelial neoplasia and cervical squamous cell carcinoma was22.22%,41.67%,70.69%. Compared between these tow groups, the expression of Ezrin in cervicalsquamous cell tissue was higher than in normal cervical tissue, the difference wassignificant (x2=13.361, P<0.05).The expression of Ezrin in cervical squamous celltissue was obviously higher than in cervical intraepithelial neoplasia organization, thedifference was significant as well(x2=6.081, P<0.05).Though the expression of Ezrinin cervical intraepithelial neoplasia tissue was higher than its expression in normalcervical tissue, but the difference was not significant, there was not statisticallysignificant (x2=1.750, P>0.05).3、The positive expression rate of CD44v6in squamous cell carcinoma of the Iand II A stage were65.71%and73.91%, there was no significant difference aftercomparing both (x2=0.436, P>0.05). In cervical squamous cell carcinoma ofwell-differentiated groups, differentiated and poorly differentiated group of positiveexpression rate as follows:42.86%,69.23%and88.89%, compared among the threegroups, the difference was significant in cervical squamous cell carcinoma poorlydifferentiated group(x2=2.637,P<0.05), and it was same in poorly differentiated andwell-differentiated group (x2=7.767, P<0.05), the difference of CD44v6indifferentiated group with well-differentiated group was statistically significant(x2=5.340, P<0.05). The positive expression rate of CD44v6in cervical squamous cellcarcinoma with lymph node metastasis and without lymph node metastasis were 94.44%and57.50%, a significant difference (x2=7.916, P<0.05).4、The positive expression rate of Ezrin in squamous cell carcinoma of I and II Astage were62.86%,82.61%, there was no significant difference after comparing both(x2=2.613, P>0.05). In cervical squamous cell carcinoma of well-differentiated groups,differentiated and poorly differentiated group of positive expression rate as follows:50.00%,65.38%and94.44%, compared between two of three respectively, there weresignificant differences in differentiated and poorly differentiated group (x2=5.115,P<0.05) and well-differentiated and poorly differentiated group (x2=8.296, P<0.05),there was no significant difference in well-differentiated and differentiated group(x2=0.897, P>0.05). The positive expression rate in cervical squamous cell carcinomawith lymph node metastasis and without lymph node metastasis were88.89%and62.50%(x2=4.172, P<0.05), the difference was significant, statistically significant.5、The positive expression of both CD44v6and Ezrin were total33cases in58cases of cervical squamous cell carcinoma, there were7cases which CD44v6waspositive and Ezrin was negative,8cases which CD44v6was negative and Ezrin waspositive, both were negative expressed were10cases, it showed that there was a highexpression of Ezrin when CD44v6was positive expressed in cervical squamous cellcarcinoma, the analysis indicated a positive correlation between them (γ=0.387, P<0.05).Conclusion:1、The expression of CD44v6in cervical squamous cell carcinoma is higher thannormal cervical tissue, suggesting that the high expression of CD44v6mightcontribute to the progress of cervical squamous cell carcinoma. The expression ofCD44v6in the poorly differentiated-cervical squamous cell carcinoma tissue wassignificantly higher, and the positive expression rate of CD44v6in the lymph nodemetastasis group was significantly higher than the metastasis group of the withoutlymph node, it was considered that the high expression of CD44v6might predicted that cervical squamous cell carcinoma had stronger ability of invasion and metastasis.2、Ezrin in squamous cell carcinoma of the cervix was overexpressed, thepositive expression was significantly higher than in cervical intraepithelial neoplasiaand normal cervical tissue, it was suggested that the high expression of Ezrin mightcontribute to the progress of cervical squamous cell carcinoma. The expression ofEzrin in poorly differentiated cervical squamous cell carcinoma was significantlyhigher, and in the pelvic lymph node metastasis, the positive expression rate of Ezrinin cervical squamous cell carcinoma might indicate with lymph node metastasis wassignificantly higher than without lymph node metastasis, and it was considered thatthe high expression might predict the degree of malignancy of tumor was high, theprognosis was poor, and easy to relapse.3、CD44v6and Ezrin were both involved in the development and metastasis ofcervical squamous cell carcinoma, the expression was positively correlated and hadsynergy in the development of cervical squamous cell carcinoma and metastasis.Combined detection of CD44v6and Ezrin expression in cervical squamous cellcarcinoma, provide a reference to determine the degree of malignancy and prognosisindicators.
