Poria cocos is the dried sclerotia of Fungus Poria of Polyporaceae, which has the role of removing dampness and promoting diuresis, invigorating spleen, calming and nourishing the heart. It is used in the treatment of edema and oliguria, unease, loose stools and diarrhea, spleen deficiency with weak digestion etc. Poria cocos is the traditional Chinese medicinal material used for both food and medicine. It has wide range of clinical applications for not only component with many other Chinese herb medicines but also as a large dose alone, treating a variety of disorders clinically. It also can be made into a large amount of healthy food with good social and economic benefits. Therefore, the deep development of Poria cocos has broad market prospects.The active ingredient of Poria cocos is Pachymaran. It has the function of anti-tumor effect, enhancing immunity, antiviral, eliminating kidney stones and can be widely used in the fields of health care, food processing. Consequently, it is a great significance for extraction and exploitation of polysaccharides. In this thesis, the main research contents include the extraction process to Pachymaran:firstly, investigating the granularity, additive water amount and the time of extraction by using ultrasonic extraction method, then on this basis, to design orthogonal experiment. To pick out the optimized ultrasonic water extraction process on the index of yield rate:The granularity is40items, the time of supersonic is30min, and additive water amount is1g:50mL. This research results provide scientific reference for the mass production of Pachymaran.Besides, because there are large and complicated chemical compositions in Chinese herb, whose original source, producing area, medicative parts and storage condition would have effect on the quality. It is quite necessary and urgent to establish Chinese herbal medicines qualitative and quantitative standards by using modern science and technology. This program also includes the research of quality control on Poria cocos:(1) Equipping the visible-ultraviolet spectrophotometry to determine the contents of pachman from different origins by sulfuric acid-phenol method. The rusults showed that the polysaccharide content is1.774%~2.422.(2) Using the HNO3-HClO4system sample digestion to determine the metal elements (Mg、Fe、Mn、Zn、Cu) of it by flame atomic absorption spectrometry.The rusults showed that Mg>Fe>Mn>Zn>Cu and recovery rate was0.487%~1.852%.(3) Adopting wet digestion method to determine the contents of Pb and Cd of Poria cocos by graphite furnace atomic absorption spectrometry. This program conducts in-depth study of Poria cocos quality assessment method through modern analysis and testing technology, so as to provide a scientific basis for Poria cocos quality control and deep development of medicinal resources.
