Poria cocos is the arid sclerote of Poria cocos (Schw.) Wolf which is a kind of mycetes belonging to Polyporaceae. Poria cocos can also be called with Futu or Songshu. Our country’s recource is rich, it is mainly produced in Hubei, Hunan,Anhui,etc. Poria cocos which come from Luotian town of Hubei province is good at quality. Poria cocos is a traditional Chinese medicine for a long history, its nature and tasting for sweet and light. Poria cocos can excrete dampness, be used as diuretic to alleviate dropsy, invigorate spleen and calm the nerves. The mainly chemical composition of Poria cocos are polysaccharides, triterpenoids, fat acid, daucosterol, Enzyme. Modern pharmacology studies have shown that Poria cocos has the flowing bio-activity:anti-tumor, regulate immune function, anti-inflammatory, anti-allergy and other aspects of bio-activity.Preliminary research of our laboratory shows that EtOAc extraction of Poria cocos has the antitumor active. This paper use interdisciplinary research methods to study on the antitumor active compositions of EtOAc extraction of Poria cocos. Using the chromatographic method to isolate and purify the EtOAc extraction of Poria cocos, four compounds were extracted and isolated. Their structures have been determined as pachymic acid(B1) dehydropachymic acid (B2)、dioscin (H1)、deltonin (H2). Dioscin (H1) and deltonin (H2) were found from Poria cocos for the first time.Taking HepG2cell、Caski cell、H460cell and MDA-MB-231cell as test objects, we screen the anti-tumor activity constituent from the above compounds by MTT assay. The results of the study show that compounds H1and H2can significantly inhibit the proliferation of hepatoma cell line HepG2、the cervical cancer cell lines caski, lung cancer cell lines of H460and breast cancer cell line MDA-MB-231, and with the dose dependent and time dependence. Inhibi-tion rate which hepatoma cell lines HepG2, lung carcinoma cell line H460, breast cancer cell line MDA-MB-231cells are dealed with by the drug’high concentration for48h and72h is over90%, especially for breast cancer cell lines MDA-MB-231. For breast cancer cell line MDA-MB-231cells, half-inhibit concentration of compound H1at24h、48h and72h are6.17μg/ml、1.82μg/ml and1.45μg/ml, half-inhibit concentration of compound H2at24h、48h and72h are1.73μg/ml、0.515μg/ml and0.171μg/ml.Through the follow method as observation of apoptosis morphological changes by Hoechst33258dyeing to explore out the apoptosis antitumor mechanism, breast cancer cell lines MDA-MB-231which was treated by compounds H1and H2respectively at the concentration of2-15μg/ml for24h showed apparent apoptosis morphological characteristics on Hoechst33258dyeing. It is very important to clarify material foundation of antitumor of poria cocos.
