Object:The present study, according to detecting pharmacodynamic effects of Scutellaria baicalensis Georgi (SBG) and Coptis chinensis Franch (CCF), a ims to explore the correlation of those chinese herbs on cholinergic anti-inflam matory pathway through the acute lung injury (ALI) model.Methods:(1) ALI was established by tail intravenous injection of lipopoly saccride (LPS). The pathological changes of pulmonary tissues, TNF-a, NO lev els in serum, protein content in alveolar lavage fluid and lung wet/dry weight ratio were tested respectively after injected with LPS1,2,4,6and10h;(2) The ALI model was established after intragastric administration SBG for a wee k and then to observe the basis situations of rats. The rectal temperature was measured after injected with LPS1,2and4h. The changes of TNF-a, NO a nd Ach levels in serum, ChAT and AChE activities in pulmonary tissues homo genate and lung wet/dry weight ratio were tested respectively;(3) The ALI mo del was established after intragastric administration CCF for a week and then t o observe the basis situations of rats. The changes of TNF-a, NO, Ach, ChAT and AChE levels or activities in serum, Ach, ChAT and AChE levels or activ ities in pulmonary tissues homogenate and brain tissues homogenate were teste d respectively after injected with LPS1,2and4h, the same as lung wet/dry weight ratio.Results:(1) Compared with the blank group, the model group could incre ase the wet/dry weight radio, TNF-a, NO levels in serum, while improve prote in content in alveolar lavage fluid significantly at1,2,4h after model set up. The pathological detected of pulmonary tissues showed that severe inflammato ry infiltration and hyperaemia;(2) Compared with the model group, the SBG showed the certain degree of protection on inflammatory response of lung tissue. SBG could reduce the e dema of lung tissue, the magnitude of body temperature changes, and the NO level in serum, while improve Ach content in serum significantly at2h after model set up. Besides, it could reduce TNF-a level in serum, but there was n o statistical significance;(3) Compared with the model group, the CCF showed the certain degree of protection on inflammatory response of lung and brain tissues. CCF could r educe the edema of lung tissue, and TNF-a, NO levels in serum, while improv e Ach content in brain tissue homogenate and the activity of ChAT in pulmon ary tissue homogenate.Conclusion:(1) The ALI model could be set up successfully via tail intra venous injection of LPS (5mg/kg);(2) SBG on cholinergic an ti-inflammatory pathway may be due to indirectl y transiently promoting the release of Ach, but not relating to the biosynthesis of Ach;(3) There were different influences of CCF on cholinergic anti-inflammator y pathway through the protective effect of ALI model. In the central system, CCF may be due to indirectly promoting the release of Ach, but not relating t o the biosynthesis of Ach; in peripheral system, it may be due to improving t he activity of ChAT in lung tissue, and promoting Ach synthesis for further re alization of anti-inflammatory effect;(4) SBG, including purging fire and detoxifying toxicosis of efficacy, has no correlation between clearing lung heat and cholinergic anti-inflammatory pat hway. Based on cholinergic anti-inflammatory pathway, CCF, including purging fire and detoxifying toxicosis of efficacy, have a certain effect of clearing lung heat.
