?Objective?In this study,the metabolic indexes of Scutellaria baicalensis-Coptis chinensis and Coptis chinensis and Coptis chinensis on type 2 diabetic mice were studied.By comparing the effects of Coptis baicalensis,Coptis chinensis and Coptis chinensis-Coptis chinensis on high fat combined with STZ to induce T2 DM fasting Blood glucose,fasting insulin,blood lipids,pancreatic NLRP3,ASC,casepase-1,IL-1?,and IL-18 expression levels were evaluated to evaluate the mechanism of protection of islet injury.?Method?1)Sixty SPF male mice were fed with high-fat diet for 8 weeks,small doses of STZ(50mg / kg)was injected into abdominal cavity rapidly for 8-10 hours of fasting.And continuous intraperitoneal injection for 3 days.Fasting blood glucose values> 11.1 mmol / L were determined to be successful.The successful mice were divided into 5 groups(n = 10)with 50 machines: model group,Coptis chinensis group,Coptis chinensis group,Coptis chinensis-Cotinus coggygria group.Another 10 SPF male mice were set as blank control group.2)Decoction preparation: Refer to the 2015 edition of Chinese Pharmacopoeia,conventional dose of Coptis chinensis: 2 ? 5g / d,compatible with Scutellaria baicalensis.The final determination of the concentration of crude drug is 20 mg / ml-50 mg / ml.In this experiment,intermediate measurement 30 mg / ml.Coptis chinensis decoction: 15 g Coptis chinensis and 2L of water for 30 minutes,and then decocted for 30 minutes to stand and condense the medicinal solution to 0.5L for later use.Scutellaria baicalensis decoction: 15 g scutellaria baicalensis pieces were immersed in 2L of water for 30 minutes,then decocted for 30 minutes,and then the solution was concentrated to 0.5L for use.Coptis chinensis-Cotinus coggygria decoction: 15 g Coptis chinensis and 15 g Cotinus chinensis decoction pieces were soaked in 2L of water for 30 minutes,and then decocted for 30 minutes to stand,and the medicinal solution was concentrated to 0.5L for use.3)Intervention: All groups were administrated intragastrically once daily at a dose of 10 ml / kg.The blank group and model group were administered sterile saline for 6 weeks.The blank group was fed daily with ordinary diet and drinking water without treatment,and the model group was fed with an equal volume of normal saline.4)During the experiment,changes in general conditions such as body weight,water consumption,mental state,and activity of mice were observed and recorded.Take fasting blood for 8-10 hours to measure fasting blood glucose;fasting insulin,triacylglycerol(TG),total cholesterol(TC),high density lipoprotein(HDL),low density lipoprotein(LDL),blood lipid,Western blot Blot(WB)was used to detect the expression of NLRP3,ASC and caspase-1 protein in pancreas.5)Statistical analysis: All survey research data in this study are recorded in the EXCEL 2006 worksheet.Statistical package for the social sciences(SPSS23.0)is used for statistical analysis of all the data,and each observation index is analyzed before and after.Contrast and comparison between groups;measurement data are expressed as mean ± standard deviation(?X±S),and if normality and homogeneity of variance are satisfied,one-way analysis of variance and group t test are used;but normality and For homogeneity of variance,non-parametric(Mann-wh1 tney U)test is used;X2 test is used for count data in two-sample classification data,and rank sum test is used for rank data.Inspection level ? = 0.05.?Results?1)Compared with the NC group,there were significant differences between the groups(p <0.05).Compared with the DM group,the level of INS in the drug group increased,and the level of INS in the DME group was the highest.This indicates that the scutellaria baicalensis-huanglian drug pair and alone can increase the INS in T2 DM mice,but the effect is not better than that of metformin and scutellaria baicalensis drug pair.The effect of increasing INS was better than that of Scutellaria baicalensis and Coptis chinensis,and the effect of scutellaria baicalensis was the second.2)Scutellaria baicalensis and Coptis chinensis can be used alone and as a medicine pair to reduce the levels of TC,TG,HDL-C in T2 DM mice,to a certain extent,to improve lipid metabolism disorders in T2 DM mice,and to reduce the effects of TC,TG,HDL-C.It is better than metformin,among which baicalone-coptis chinensis has the best effect,followed by baicalone alone,and baicalone alone is the worst.3)Compared with the NC group,the levels of IL-1? and IL-18 were significantly different between the groups(p <0.05),which was statistically significant.Compared with the MC group,the levels of IL-1? and IL-18 in the drug group were significantly reduced.Metformin,the baicalin-coptis chinensis drug pair,and the baicale coptis and berberine alone could reduce the levels of inflammatory factors IL-1? and IL-18 in T2 DM mice.The DCS group had the largest reduction,better than the DME group,followed by the DC group,and the DS group had the worst.This shows that the baicale-coptis chinensis medicine pair and its use alone can improve the level of inflammatory factors in T2 DM mice,and the baicale-coptis chinensis medicine is better than metformin.4)Scutellaria baicalensis-Coptis chinensis pair and single use can down-regulate the expression of NLRP3,caspase-1,and ASC protein in the tissues of the pancreas,and the effect of scutellaria baicalensis-Pinus baicalensis is better than metformin,and the effect of baical or berberine alone is not better than metformin.5)Scutellaria baicalensis-Coptis chinensis pair and single use can down-regulate the expression of NLRP3,caspase-1,and ASC protein in the tissues of the pancreas,and the effect of scutellaria baicalensis-Coptis chinensis on Metformin is better than metformin.?Conclusion?1)Scutellaria baicalensis-Coptis chinensis can effectively improve the symptoms of experimental diabetic mice,reduce blood glucose,and promote insulin secretion in diabetic mice.Scutellaria baicalensis-Coptis chinensis is better than scutellaria baicalensis and Coptis chinensis in reducing glucose and improving insulin secretion.When used alone,the Coptis chinensis group had a better blood glucose lowering effect than the Cotinus coggygria.2)The scutellaria baicalensis-coptis chinensis combination and its single use can improve the lipid metabolism disorder in experimental diabetic mice.The scutellaria baicalensis-coptis chinensis is better than scutellaria baicalensis and Coptis chinensis in improving lipid metabolism,and the scutellaria baicalensis improves lipid metabolism better than the scutellariae group.3)Scutellaria baicalensis-Coptis chinensis medicament and monotherapy can decrease the levels of inflammatory factors in pancreatic tissues,which may inhibit the activation of NLRP3,improve pancreatic tissue damage and protect islet function.The scutellaria baicalensis-coptis chinensis medicine has the most obvious effect on improving and improving pancreatic tissue damage.Coptis chinensis is better than scutellaria baicalensis alone.
