Yalian originates from Coptis deltoidea C. Y. Cheng et Hsiao rhizome, with Qingre dampness, Xiehuo detoxification function, famous for production of genuine medicinal materials in. At present, Chinese Pharmacopoeia2010edition of Yalian alkaloid limit lack, affected the quality control of Chinese medicinal materials. In addition, soil factors also affect Yalian chemical components and quality of one of the important factors. Therefore, to carry out Yalian fingerprint and establishment of quality standard, chemical composition and soil factor correlation study, to ensure the quality of medicinal materials Yalian and clinical drug efficacy has important significance.Objective:Based on the resource survey as the basis, to carry out Yalian chemical composition analysis, establishing fingerprint and quality control system, and the study of Yalian of soil factor and medicinal correlation between chemical composition, as Yalian resource conservation, cultivation and quality control to provide the scientific basis.Methods:1. By sampling, investigation methods, even feel elegant resource distribution, folk custom goods and product confused situation;2. Using the method of HPLC-DAD, determination of berberine type alkaloids in6species of Yalian component content, and the establishment of different origin Yalian chemical fingerprints.Using the method of1H-NMR, the establishment of different origin Yalian1H-NMR fingerprint;3. Using pH and STAL-2nitrogen phosphorus and potassium in soil determinatio n instrument, determination of the soil pH, organic matter, ammonia nitrogen, available phosphorus, available potassium content;4. By the method of grey system analysis Yalian chemical composition and soil factors. Results:1. Yalian resources are mainly distributed in the eastern margin of the Tibetan Plateau in Qionglai mountains area, cultivated mainly in elevation between1100-1400m; wild products are mainly distributed in the elevation of1400-1700m.2. Based on the characters of Yalian, authentication, inspection, thin-layer chromatography, content determination and chemical composition analysis, establish a standard Yalian quality standards, norms Pharmacopoeia Yalian berberine type alkaloids limited.3. Detection of the17batch of Yalian samples of HPLC-DAD fingerprinting, obtained12common peaks, the calibration of magnoflorine and other7kinds of alkaloids.Fingerprint similarity was above0.97, indicating different origin with minor differences; folk custom products fingerprint and Yalian has higher similarity; preliminary identification of adulterant species may even to taste.4. Established Yalian’H-NMR fingerprinting, and HPLC-DAD fingerprints were compared and analyzed, the Yalian secondary metabolite fingerprinting and metabolite fingerprinting has certain difference.5. Yalian chemical composition and soil factor correlations indicated:soil organic matter is the five factor to Yalia chemical composition affect the smallest element. Among them, the correlation coefficient is greater than0.7200can be:and magnoflorine related soil are main factors pH and ammonium nitrogen;6kinds of berberine type alkaloids and related main soil factors are available potassium, ammonium nitrogen and available phosphorus. Magnoflorine and berberine type alkaloids in6species showed different results, the reason is that the two alkaloids have a different mother nuclear structure, while the different chemical structure formation may be affected by different biosynthetic pathways of influence, different biosynthetic pathway required for soil factors also may be different.6kinds of alkaloids of three main factor of soil available potassium, with the highest correlation, this and "ICP-MS simultaneous determination of different origin of the contents of33trace elements in the article of a study on the determination of trace elements in Yalian consistent results, Yalian medicines containing relatively high K element.Conclusion:Yalian medicinal materials in the chemical composition and soil factors of the comprehensive evaluation, the pure component analysis can not effectively control the quality of medicines, adopting various analysis means, establish "zoology-Chemistry" correlation analysis, may be improved Yalian quality control approach.
