Turnip is the radix and seed of Brassica rapa L.. The drug is collected in autumn when the radix and seed are ripe. Turnip is also called NIUMA (from Tibetan medicine’s classics Four-Volume Medical Code) in Tibetan herb, and is generally called MANJING, YUANGEN in Tibet. In traditional Tibetan Medicine theory, turnip has the effect of detoxification and nourishing. It is suitable for treatment of all kinds of poisons.’Long’ illness and body weakness. At present, there are no reports about the screening of antifatigue and antioxidation of turnip, and the simultaneous determination of quercetin, kaempferol and isorhamnetin in turnip leaves by HPLC.PurposeTo screening the antifatigue and antioxidant biological activity, and establish the quality analysis methods for turnip in order to provide the evidence for health food developing on turnip.Methods1. Using the mice swimming experiment to screening antifatigue activity of turnip;2. Using the DPPH free radical scavenging experiment to screening antioxidant activity of turnip;3. Using the phenol-sulfuric acid method to determine the total polysaccharide content of turnip;4. Using the TLC method to identify quercetin and isorhamnetin of turnip leaves;5. Using the HPLC method to determine the contents of quercetin, kaempferol and isorhamnetin of turnip leaves.Results1. The water extract of turnip has the significant antifatigue effect in the range of0.38-37.62g(turnip herb)/kg/times,and the dosage of0.38g (turnip herb)/kg/times has the best effect in all dosages. The crude polysaccharide of turnip has the significant antifatigue effect in the range of0.33~0.59g (turnip herb)/kg/times. 2. The IC50of the clearance rate of14batches turnip on DPPH·is in the ranges of11.7483mg/mL~98.9100mg/mL, and the IC50of Vc, quercetin, kaempferol and isorhamnetin is0.0252,0.0404,0.0457mg/mL respectively.3. The total polysaccharide of6batches turnip is in the range of17.22%30.89%.4. Establishing the HPLC determination method for quercetin, kaempferol and isorhamnetin in turnip leaves, the their average content in3batches turnip leaves are0.01%,0.11%and0.03%respectively. The TLC identification method of quercetin and isorhamnetin in turnip leaves by TLC had been established.Conclusions1. The water extract and crude polysaccharide of turnip have the antifatigue activity and the methanol extract of turnip has good antioxidant activity in vitro. The results can be used as experiment basis for related product development.2. The established content determination for crude polysaccharide of turnip, for tree flavonoids in turnip leaves and TLC identification method for quercetin and isorhamnetin in turnip leaves, can be used as experiment basis for establishing the quality standard of turnip.
