Objective:Comparative analysis the expression level of tumor stem cells markers such as CD133, CD44, Musashil, Musashi2, OCT4, Nanog, Sox2, Bmi-1gene in biopsy samples of fiberoptic bronchoscopy(FB) in patients with lung cancer, and research whether the diagnostic value exists in Cancer stem cells in lung cancer,then to explore which stem cell markers could be served as novel molecular for human lung cancer.and evaluate its diagnostic value of lung cancer. Methods:Reverse transcription polymerase chain reaction (RT-PCR) was used to examine every gene for biopsy samples of FB in91patients with lung cancer and27patients with benign pulmonary disease.Results:①Musashi1, Nanog, Musashi2, in the expression of lung cancer significantly higher than the control group(P<0.05), the expression of Bmil, Oct4, Sox2, CD133, CD44in the experimental group and control group is no significant difference (P>0.05);②The specificity and sensitivity of Musashi1in lung cancer was highest (P<0.05).(3)No significant correlations were found between the expression of Musashi1and clinical pathological indexes including age, gender, smoking, occupation,the tumor size and pathological types of cancer(P>0.05).④The gray degree ratios of the Musashi1mRNA belt to the β-actin mRNA belt varied accordingly to different pathologic grades and degree of differentiation of cancer.There were significant difference between them(P<0.05). The positive correlation existed between the expression of Musashi1mRNA (r2=0.94, P1=0.027)(R2=0.90, P2=0.004)Conclusions:①Musashi1, Nanog, Musashi2can as lung cancer markers of cancer stem cells express in human lung cancer organization; Sox2, CD133, CD44, Oct4, Bmi1will not be as lung cancer markers of cancer stem cells there, and Musashi1, Nanog, Musashi2in the expression of lung cancer significantly higher than the control group.②The specificity and sensitivity of Musashil in lung cancer was highest, and the positive correlation existed between the expression of Musashi1mRNA, So Musashi1in the human lung cancer can be used in diagnosing judging the malignancy and the prognosis.③Detecting TSC mRNA as molecular pathology in the human lung cancer would retrieve the limitations of traditional tissue pathology.
