Meyer are greatly supplement original qi,restore the pulse and stem desertion which are treated as heart failure and cardiogenic shock in clinical application.Pure Ginseng Decoction was firstly record in A Miraculous Book of Ten Prescriptions by Ge Kejiu in Yuan period.Ginseng powder was ancestral prescription of Pure Ginseng Decoction which was record in A Handbook of Prescriptions for Emergencies by Ge Hong in Jin period1600years ago. Pure Ginseng Decoction can not be replaced by other drugs and it’s a first-aid prescriptions in TCM clinic.We set out from clinic of TCM to establish a Pharmacology experiment method which is fit Pure Ginseng Decoction in Medication features.To screen the effective components is the point of this paper.The paper set up experimental heart failure model on rats and the route of administration of Pure Ginseng Decoction is in duodenum.To observe the index of hemodynamics which are HR, LVSP,MAP,LVEDP and+dp/dtmax,we found the6g.kg-1dosage had better effects than3g.kg-1dosage. The6g.kg-1dosage made raise in varying degrees in30-40min of HR,LVSP,MAP,+dp/dtmax and restrain LVEDP which was raise on model rats.Also we carry through the Maximum tolerated dose (MTD) and it showed that the mice were all survival with maximum concentration(1.5g-mL-1) and maximum volume(0.04mL-g-1) after7days’observations.The administration dosage was400～1200times than a daily dosage(3～9g).Through the experiment we proved the safety in using megadose of ginseng.In order to study the effective components of strengthening heart effects in Pure Ginseng Decoction,we used system solvent extraction and extracted petroleum ether extract, ethyl acetate extract, n-butyl alcohol extract and water extract. The acute heart failure model were used to study the four extracts respectively to screen the strengthening heart components ulteriorly.The results indicated petroleum ether extract and ethyl acetate extract that had declining tendency but no statistical significance(P>0.05). There were strengthening heart effects observably with n-butyl alcohol extract(P<0.05).The experiment of water extract showed strengthening heart tendency but no statistical significance(P>0.05).By reading references and our experiment,the main component in ginseng had effects of strengthening heart are ginsenosides.So we chose9ginsenosides(Rg1,Re,Rf, Rg2, Rb1, Rc, Rb2, Rb3, Rd) as standards to detected the content with HPLC and the results respectively were5.18mg-g-1,4.54mg-g-1,1.72mg-g-1,0.74mg·g-1,10.55mg·g-1,10.61mg·g-1,7.14mg·g-1,0.08mg·g-1,4.68mg·g-1.It’s reported that total flavonoids of ginseng had effects of lower blood pressure.In order to discuss components in ethyl acetate extract which might have strengthening heart effects,we detected content of total flavonoids with UV and the content was0.15mg·g-1.Through our experiment,the water extract had the tendency of strengthening heart,so we detected content of total polysaccharides with UV and the content was359.90mg·g-1According to the analysis of the experiment,6g·kg-1dosage of Pure Ginseng Decoction can improved the index of hemodynamics to enhance the heart working in a short time.The MTD results proved the feasibility of using large doses of ginseng for theory basis in rescuing of acute heart failure.The main strengthening heart effects component of ginseng is n-butyl alcohol extract.The water extract showed unconspicuous effects but can ameliorate the index of hemodynamics. Although petroleum ether extract and ethyl acetate extract had no strengthening heart effects,we can eliminate the interferential components to enhance the positive effects.All these we intend to do more experiments next step.
