In this paper,the main ginsenosides of raw ginsengs were studied and analyzed;and the convertion from the protopanaxadiol type ginsenoside(PPD type ginsenoside)to Compound-K(C-K)by enzyme was also studied;then,to analyze rare ginsenosides in the red ginseng products and Chinese traditional drugs(Zhongcaoyao),the analysis method of the 16 kinds of red ginseng rare ginsenosides was established;finally,the rare ginsenosides in red ginseng products such as red ginseng slices,red ginseng extract,and in the Chinese traditional drugs containing ginseng were studied and analyzed.Firstly,the ginsenosides content and the different ginsenoside proportion in the ginseng,american-ginseng and notogingseng root and fiber were studied and analyzed.The content of total ginsenoside in the ginseng fiber and american-ginseng fiber was 1.5 time of that in the root;however,the total ginsenoside content in the notoginseng root was 3 times of that in the fiber.In the ginseng,the content of main ginsenoside such as Rb1,Rb2,Rc,Rd,Re and Rg1 was similar;and the content of ginsenoside Rf was 5-8%of the total ginsenoside as a special ginsenoside,and the ginseng also contained a little of ginsenoside R1.In american-ginseng,the ginsenoside Re and Rb1 contents were higher,over 40-50%of the total ginsenoside.In the notoginseng,the different ginsenoside proportion in the root and fiber was similar;the higher content ginsenoside was Rg1 and Rb1 which contet was over 70%in the total ginenoside;content of ginsenoside R1 was 9-10%as a special ginsenoside.These are very usable for developing new ginseng products selecting raw materials of ginseng kind.After oral administration of ginseng,the PPD type ginsenosides were converted to C-K and then absorbed by digestive system to play efficacy.To obtain C-K,the convertion PPD ginsenoside to C-K was studied.The good enzyme production was obtained in low-cost medium by the strain sp.848;the optimal substrate concetration(PPD ginsenoside)of enzyme reaction was 7%,the optimal temperature was 50 ?,the reaction time was 22 h,the optimal pH was 5.0.And the conversion rate of C-K was over 80%.To analyze rare ginsenosides in ginseng products,the UPLC-MS method was used to analyze rare ginsenosides of the red ginseng such as 20(S)-Rg3,20(R)-Rg3,Rgs,Rk1,20(S)-Rh2,20(R)-Rh2,Rh3,Rk2;20(S)-Rh1,20(R)-Rh1,Rh1,Rk3;20(S)-Rg2,20(R)-Rg2,Rg4(F4)and Rg6 to made a foundation for identification and analysis of rare ginsenosides in the red ginseng slices and red ginseng extract.The transformation of ginsenosides in the red ginseng products such as red ginseng slices were analyzed and compared.The content of ginsenosides in red ginseng extracts were analyzed.In the newly produced red ginseng slice by using the ginseng oneself ginsenosidases,the content of Re and Rgi was similar with the american-ginseng level,and the main ginsenoside Rbi,Rb2,Rc,Rd,Re and Rgi were tranged to rare ginsenoside 20(S)-Rg2,20(R)-Rg2,F2,20(S)-Rg3,20(R)-Rg3,Rg5,Rk1 C-K,20(S)-Rh2,20(R)-Rh2,Rh3,and Rk2,which content was higher than that of traditional red ginseng.In the red ginseng extract,the analysis results showed that during the processing of extracting red ginseng,the main ginsenosides Rbi,Rb2,Rc,Rd,Re and Rg1,were mostly transformed to rare ginsenosides such as 20(S)-Rg3,20(R)-Rg3,Rg5,Rk1,20(S)-Rh2,20(R)-Rh2,Rh3,Rk2,20(S)-Rhi,20(R)-Rh1,Rh4,Rk3,20(S)-Rg2,20(R)-Rg2,Rg4(F4)and Rg6,which were easily absorbed and had higher activity.A high-performance liquid chromatography(HPLC)method was developed and validated for the simultaneous determination of fourteen ginsenosides such as Rgi,Rf,Rbi,Rb2,Rc,Rg2,Rd,F2.Rh4,Rk3,20(S)-Rg3,20(R)-Rg3 and Rki(Rg5)containing in the traditional Chinese medicine:Yi-Qi-Fu-Mai injection.During analyzing,it was used six concentrations for every ginsenoside,and each one was detected three times.In the calibration curves of ginsenosides,the r2 values above 0.9990 for all the analytes,and the good linear relationships were observed.In this method,the precision,robustness and stability were recognized.The average recoveries for the eleven ginsenosides was 96.21%.This method was found to be rapid and accurate,it could be readily applied to the quality assessment of Yi-Qi-Fu-Mai injection and other traditional Chinese drugs containing ginseng.
