In this paper,cultivation, Microscopical identification,content of Gastrodin in different growth stages and different organizations, GC-MS analysis of petroleum ether extract,chemical composition of sterms of Gastrodia elata B1.f.glauea S.Chow were studied.On cultivation, undergrowth bionic cultivation technology of Gastrodia elata B1.f.glauea S.Chow from Changbai Mountain has been studied,mainly studied the preparation of Gastrodia elata B1.f.glauea S.Chow germination and symbiotic bacteria, and the cultivation method for sexual propagation and asexual propagation which could adapt to Changbai Mountain area. Experiments show that by using this method,the breeding method of Gastrodia elata B1.f.glauea S.Chow is short,low risk,strong tubers, the drying rate is around10%, and the final product of Gastrodia is solid,good quality of appearance,and less number of wrinkles which are not obvious.Using this method,input25RMB can produce2.0kg commodity Gastrodia elata B1.f.glauea S.Chow and4.0kg seed Gastrodia elata Bl.f.glauea S.Chow,the input-output ratio is1to5.On Microscopical identification,the microscopic structure of the different growth stages of Gastrodia elata B1.f.glauea S.Chow and flowers were descriped though the hand-sliced method and paraffin method,there are no significant differences among the cross-sectional slices of sword gastrodia、white gastrodia and mother Gastrodia except the vascular bundle,Under the skin all by bolts of cells,under the subcutaneous for cortical organization,the next is the thin-walled cells, thin-walled cells nearly colorless,thin cell walls, the nucleus and cytoplasm were seen in the thin-walled cells,some thin-walled cells also contain calcium oxalate crystal beam. vascular bundles distribute at the column layer, spiral duct can clearly seen in the longitudinal section of vascular bundles,the vascular bundles of mother Gastrodia are different from the mature tuber and the immature tuber, cells containing pale yellow material, and the surrounding cell walls are thicker,Overall, tuber cells of Gastrodia elata B1.f.glauea S.Chow is gradually increasing from outside to inside, and the corresponding structures of mother gastrodia is bigger than the sword gastrodia, the corresponding structures of sword gastrodia is bigger than the white gastrodia.On chemical compositions,by using silica gel column chromatography and thin layer chromatography,4chemical compounds are got from the different extract of Gastrodia elata B1.f.glauea S.Chow stems, and chemical construction indentify is carried out by using13C-NMR,1H-NMR, DEPT, ESI and other devices. The5chemical compounds are Mannitol, p-hydroxybenzoic acid, β-sitosterol,1,2-O-Isopropylidene-D-glucofuranose, Trimethylacetic acid. GC-MS analysis is carried out for petroleum ether extract of Gastrodia elata Bl.f.glauea S.Chow at different growth stages and different parts:using WIKEY.L. map retrieval, combined with base peak,load ratio and the relative abundance of mass spectrogram to standard spectrogram,indentifies31chemical compounds from sword gastrodia,30chemical compounds from white gastrodia,11chemical compounds from mother Gastrodia,31chemical compounds from seed Gastrodia,7chemical compounds from stalks and20chemical compounds from flowers.Using the testing method of Gastrodia glycoside content in Phamacopoeia to determine the Gastrodia glycoside content in different growth stage and different orgnizations.The results show that Gastrodia glycoside content from high to low order is:sword gastrodia> white gastrodia> mother Gastrodia>stalks>seed Gastrodia.
